Inoculation Loops

So this is my loop... Ya pretty ghetto! I hate these wire loops and they didn't come with a holder, fer cryin out loud! And the wire is too flimsy, it just bends at the slightest touch.
So, I searched the Shroomery, no joy...
So, this POS loop is .010 wire diameter, 30 AWG, and loop is .160 ID or 4 mm Mostly I see 24 and 25 AWG offered.
μL, what the hell is that μL? mi·cro·li·ter /ˈmīkrōlēdər/ one millionth of a liter. loop Volume One μL loopful of urine!
Loop diameters: 3mm =.118, 4mm=.157, 5mm=.196 1 μL to 10 μL volume of piss!
In 1960 O'Sullivan published a study carried out in Birmingham comparing the colony counts obtained with the pour-plate method with those obtained by culture of a "standard loopful" of urine. For the inoculating loop method, a platinum wire loop , with 3 mm internal diameter, was used to sample the urine specimen, and the loopful spread over a nutrient agar plate. more article... The standard loops are made from twisted Nichrome 5 wire. Nichrome 5 is fast cooling and exceptionally durable, and the twisted shank does not vibrate as much as single wires.
I'm not a heavy user of the loop, in fact I haven't even put a new one in. But on the other hand I'd like to try different wire gauges. A 4 mm loop seems fine.
So what do you use? 1. A loop holder with fixed loop? 2. A loop holder with replaceable loops? 2.1 Yer own ghetto wooden dowel with the loop stuck in the end? 3. 25 AWG, 26, 30? 4. 3mm dia., 4, 5, loop? 5. A pretty pink loop holder cause yer such a girly girl! 6. An Exacto holder with Kenthal Vape Wire 7. Stripped TV cable wire even though you own a $500 AA PC! LOL 8. Just hold the loop with yer fingers!

I use the same ones you have but it’s such a hassle getting them in the handle I started just holding them.
Never had a problem with them bending, I just rub the different sides of the circle on the print. It helps if you cool the loop in agar then rub the print quick and they get glued to the agar.

I made my own with electrical wire (unknown gauge). It's a little too thick though. Takes a lot of flaming to get red hot.

I made one with some nichrome wire and an Xacto handle. I just stuck the two ends of the wire in the handle then wrapped it around a toothpick. I still haven't replaced it, it's very durable

I use qtips as swabs. Individually wrapped in foil, put in a jar, pressure cooked 15-20 mins.

Kenthal vape wire. And just use an xacto handle, insert wire, tighten collet, done.

I typically use 10ul disposable plastic pre sterilized loops. Theyre cheap and easy, no need to sterilize. Blah blah blha
Recently I bought a cheap $7 loop handle with 50 tips. Really I just bought it for the clamp handle. I like a stiffer loop too, and these wire loops are fairly flimsy (though thicker than yours by the looks of it). I plan to make my own loop tips with all the vape wire I have sitting around from the year I quit smoking and started vaping. Ive got oodles of spools of wire. Kanthal, NiChrome, Stainless Steal, etc.

Oh and uL, I think thats how much water the loop could hold. Its a "micro" unit, which is a millionth, so a millionth of a liter. So a 1uL loop would take a million dips to transfer a liter of water over.
At least thats my best guess from common sense thinking. Could be wrong.

there is a pottery tool for every myco use. stored with clay, your tool kit becomes invisible.

Stripped a piece of typical TV cable, fairly thick piece of copper wire, some fancy bends, used exacto for a handle.

I'm gonna have to go out to the garage and strip me some TV cable LadysKnight! I'll wrap around a .157~ drill for a 4mm loop.

Rocking the inoculating loop he made ripping cords off his tv, but has a shiny $500 AA PC on the floor.
