Here we are again. Another step in this beautiful adventure we call life. Being a human. Again it's been a while but im back for another update. After something like 200 days i spawned the jars that had been fully colonized for over a month to two shoeboxes of verm only :freewilly: cause why not and against everyone's suggestions here and at first, i was skeptical but they are on their second flush and no green monster yet! I had mistook some bruising early on for mold but twas a mistake.One tub is putting up creamy colored fruits with whiter gills then normal. The fruit still bruises blue and still provides a nice experience though :smile: Anyway here's where we're at!

First flush:

As i heard pasty once say its nothing to write home about or anything but hey fruit in hand while i start up some other projects from scratch :stoned:
I moved to a new place and have a whole space to myself and my antics again :hehehe: and i got tired of wasting time and spores so i quit fucking around and switched to pour plates so i could get the fuck to work. Thank you so much c10 for putting me on the right agar path your directions on your tek mixed with D3monic's perfect transfer tek has got me so ready to do some SAB work!







Here's the majority of my MS-> Grain jars just so i can get started asap and get prints out of the ones that don't contaminate. hopefully, spore works makes a nice clean syringe :thumbup:

Also since im sure some these wont make it to a shoebox i went ahead and broke into one early since i hate starting cultures with a syringe on agar and went ahead and took some APE colonized grains and put them on agar to start working faster. I basically used a jar as a germ plate which is a waste of time and resources but hey, it was just a pint.

I also got into tissue cloning. I took a "biopsy" from the center thick fruit (the 1 in the ball jar suspended in water in the picture above) and put it onto a few dishes. As well as a couple of pins from a cluster on a couple of dishes.  Hopefully That goes well first time experimenting with that.

I'm in way over my head for just starting real agar. I have 150x petries and i can already see how thats not going to last when i just started so many projects. My APE, AA+, PE6, my PE6 pin clones, My PE6 Chunk clones, my TC like jeez. Im gunna need more Petris. :lolsy:

Anyway thats the update so far. Ill be back sooner than last time, promise to anyone who cares enough to follow along at this point.  :fingerpistol:

One box got about 3 different types of mold post second flush. The other box is completely mold free and just pushed out the third what im calling her final flush. Pleasure doing business with ya time to get a new project in the shoebox.

Glad you liked it man! I just re-read it myself. It really taught me a lot committing to writing stuff out and I've come so far since last year it feels. About to have to switch to monos when all this grain spawn is ready. Im not keeping up with 50 shoeboxes

Well this sucks I definitely thought I posted earlier this morning but I must not have hit send anyway.... Ababypheonix, i’ve got to say I don’t even know you but I feel like a proud father see how far you’ve come and not only how much you were able to accomplish but also documenting it as thoroughly as you did as well. Kudos... keep it up!! You’ve definitely got quite a following! I wanted to ask you by the way I’m not sure that if you mentioned it at all throughout your documenting but what kind of agar do you use and what do you run your nutrients around do you run them low or normal? I myself have just gotten to the point where I’m feeling very comfortable with making my own agar/cleaning cultures/ identifying contam. Just recently I had transferred a sector From PDA to MEAY and After eight being in shock for about three or four days Begin to grow extremely aggressively and rapidly.... I was actually admiring your pics and couldn’t stop reading so I had dropped an agar wedge in my sab instead of into the grain jar lmao... at least I got 1 in!!


Here’s the monster ( obviously it needs to be cleaned up more but that was just a 2nd transfer after germination)




And this is never a good thing....

Well some tips that come to mind from your pics, take smaller transfers. RR recommends a grain of rice size transfer or smaller to minimize genetics and get closer to an isolate and away from possible hidden contams faster.

I use a a couple different agar techniques depending on what I'm trying to accomplish with my agar. For my standard agar plates i use C10s recipe and prep routine with D3monic's transfer tek using the leather punch hole to punch out transfers which becomes really efficient when you're doing 20+ transfers off a stack. Just flame the punch and go to town on your transfer punches takes a minute instead of cutting each one out increasing time you have the dish open. I can make 8 punches off a dish in under 10 seconds moving slowly ofc reducing contam rate i do believe.

My chitwan clone one transfer from a cloned fruit from open air grow So my recipe is

1000ml water (i use tap and never had issues for agar)
20g agar
14-15g Nute (LME)

And he has what brand he uses in the writeup (d3's) i use the same. When i want to stress a culture and get test rhizo ill run a lower nute content like 14, a bit below standard. When I'm making plates that will be the last transfer until grain (and i want to do LI) i make them with less agar so when i put the puck into my liquid inoculant it breaks up easier. I also make them gold to remind me they are good to go and hopefully to use them before they eat all the coloring lol, but this is all Just preference though.

Awesome thanks and I will... and BTW I hope you didn’t think what I had in my hand was intended for an A2A transfer lol that wouldn’t be helpful one bit lol.... and I usually try to get them as small as possible, if I can see I definitive strong sector that looks like it’s JUST starting to branch off ( and is the fastest growing out of the rest) I’ll try to grab that with just a LITTLE extra agar as to not disrupt the uniform growth. But again keep up the awesome work, ur patients and diligence has clearly paid off! Thanks and good luck

EDIT: I now see what you’re referring to when you mentioned taking smaller samples.... during those first few days of shock I wasn’t sure if I either had some contam going on or if it was just reacting unpredictably due to the yeast but it wasn’t looking good at all... plus I foolishly didn’t add enough agar at the time and it was a very gelatinous texture more along the desired consistency for germinating spores sooo a variation of those factors led to me not being able to get a clean small Xfer




As you can tell from this pic I attempted to take the transfers probably 1 day maybe 2 before that pic, you can notice that it went from being almost 100% tomatose looking to a very suspicious rizo.... had I known what it was evolving into at the time I would have gladly waited but that’s what I love about this hobby.... you can never know it all really ✌🏽👍🏼
