Dry tissue rehydration in peroxide and bleach
    #26606547 - 04/16/20 07:44 PM (3 years, 9 months ago)

I am testing various ways of cloning dry mushroom tissue, but without much success due to immediate contamination.

Tried various combinations using agar plates:
- rehydrating in 3% peroxide
- 10% bleach
- dry tissue
- used mush cap and stem samples

I am using laminar hood and always test the agar plates by keeping one plate empty. So I know it is not the environment, but the tissue itself.

Contamination develops with every method I tried, usually within 2 days.

The only thing I did not try is using antibiotic agar.

If you have experience with this, I would love to hear your thoughts.

Mushrooms are fruited in open air and dehydrated in an even more open environment. Your gonna get healthy mold spores that will always outrun your dry culture.

I have tried this it doesn't work. Sorry to break the bad news.

Oh, really? I have been planning to try cloning p. serbica this way... Is it widely accepted that "dry cloning" doesn't work? I thought that cronicr had some success with it?

Do you rip it open and pull a tiny smile from the inside? After washing the outside with alcohol

Exactly one person ever maybe.
I'm sure more have done it but it's absolutely not worth the time unless it's the last sample of a near extinct species.

Or if you're just having some time on your hand and want to have some fun.

I suppose. But I'd rather use all my time and supplies making things that work and are a good use of the time and resources
