I put absolutely nothing in this jar...What in the hell is this?
    #26587683 - 04/08/20 11:48 PM (3 years, 10 months ago)

So OBVIOUSLY there is contamination but the bottom parts of the jar are indistinguishable from mycelium it even is consolidating I put two agar wedges in two grain jars, so im thinking i grabbed the wrong jars out of two i inoculated but all three jars have white growth in them

Looks like mold. You need to sterilize your jars for longer.

If u put absolutely nothing in the the jars id evaluate your grain prep.. If u put agar wedges in i would evaluate sterile techniques..

I did them for a pretty long time I think, theyve been sitting though. I cant tell the difference between the bottom growth and mycelium, middle and above is easy.

Im going to assume its grain prep goddamn I hate that the jar is that messed up, I did pf tek and my first attempt went smooth, so I was holding myself to a high standard. I suppose it could be my sterile technique too, but I felt it wasnt that bad though I was literally sweating, outside of my sab. I dont know if my agar was screwed up either or if I grabbed a bad wedge, but yeah I PUT NOTHING IN HERE, I dont think.

May also be a filter problem

No I ditched them (2) they had mold in them, I figured, if i took from the middle of mycelium growth then thered be no contams, first time with agar though so I may learn quick enough

I’m no agar guy but my understanding is that you have to transfer out multiple times to clean up the culture if there’s sporulating mold in it.. that tissue you took was probably loaded with mold spores floating around on the plate... but I could be wrong

Like on top of it? Holy shit im a dumbass. Man but shit is weird af for me right now though, I have three jars which all look like they have mycelium but I only did 2, Theres one jar that looks pretty good (the one I want to g2g) but it actually looks less like it has actual mycelium than the other two. And i think the one that I didnt even touch has thick white growth in it, it does have obvious contamination but on the bottom it kind of looks like rhizo growth, and its consolidated and pulled away from the sides of the jar. Sorry I know this wasnt the question I asked.

Ok... so one jar you didn’t inoculate but it somehow became knocked up with what looks like myc growth?  Like immaculate conception? Lol  Some molds’ growth looks like mushroom mycelium.. or it could be another fungus, but what it’s probably not, is cubensis... unfortunately.  Whether it is or not, the rest of the jar is fucked anyway and going to be no good to you

If it was unopened then its more likely to be a lid or filter problem. Mold dies off pretty easily we sterilize for longer to deal with bacteria and endospores.

Micropore tape?
I guess alot of people use it as a filter and get away with it but there are some pics somewhere on this site of the pores in micropore tape under a microscope that show the pore size is actually too big to be able to filter mold spores. Did you use a single layer or double it up.

Man I shouldve thought about doing that, just one layer. Thatd make alot of sense if that was the problem.
