Flow hood and agar question?
    #26579382 - 04/05/20 12:16 PM (3 years, 10 months ago)

A simple question, build myself a flow hood and today is the first time using it with agar.

Does my flow hood need to be working (turned on), while the agar is cooling off (agar already poured into the dishes)?

Cheers Honey

Oh and one more thing, if I've poured some extra dishes that I do not need straight away, do i have to parafilm it before storing them in a zip log bag, or can then just go in without the parafilm?

Cheers Honey

I don't think so, I turn mine off once I got everything poured.

What I do when I'm pouring sleeves is I open the sleeve from the bottom, stack the plates 2 high (If you pour too hot and your plates warp, ur kinda fucked if you have more than 3 plates stacked IME). and keep the plastic so when everything is solid, you can stack the plates and put the plastic sleeve back over: easy peasy.

Yeah? But do you keep the flow hood on when the plates are cooling or no?

Personally the labs I've worked in have had the resources to keep flowhoods on longer (funds for replacing filters and the electricity to keep them on) so I suppose the answer is yes? It's not entirely necessary though. I've poured agar (pretty near boiling, mind you) into plates out in the open and they didn't contaminate, no air filtration, no nothing just boiled agar and poured it.  I personally do no-pour in an SAB because I haven't bitten the bullet and built a flowhood yet.

I would always leave it on until they had parafilm round them. So pour, cool, and seal with flowhood on, but it was an old house with several mouldy windowsills so I was over cautious. I'm also clumsy, the sound of the fan would remind me that they were there and prevent me knocking them everywhere.
In regards to storing them, always seal them before storage. The lids can easily become displaced and compromise their sterility.

I use ziplocks, only. 1 per plate.

Leave the hood running til they cool and you package them.

I recommend pouring a whole sleeve at once. They will keep for a couple of months no problem.

I put them 3 to a sandwich bag with no film. I put them in the bags in front of the hood and when I go to use them I pull them out in front of the hood. They get film after inoculation.

I go through about 10 dishes a week using this method. Never any issues.

Also if you search Amazon you can buy a case of 500 petri dishes for about $70. Just an FYI. They're good plates too
