First Agar with pics UPDATED

I poured some plates, 20g agar, 15g lme, 1 liter h20, in SAB.

I streaked them with a loop from a print I made 5 years ago from a B+ strain.

They all look fairly similar in growth, I only see 1 spot with confirmed contam(small yellow spot in pic 2).

I am wondering when I should start pulling transfers to new dishes, could I take one whole growth area that looks isolated like the spot on the 3rd pic, or should I stick to leading edges? This is my first time doing agar and I haven't done any transfers yet, just pours and streaks from prints.

They need to grow a little more to be sure and to be ready for transfers
post again in about a week and we'll be able to tell better

My rule is that sooner is better when transferring to reduce contams setting up on the dish and spreading. You have to balance this against getting good sectoring and your comfort level with wielding the scalpel when dealing with small wedges.

You could definitely take the isolated growth on #3 and move to a new dish so you get a single colony. From the new dish, take a transfer from the leading edge when ready. The final dish should be pretty clean by now. Observe and decide from there whether to clean up some more with additional transfers or move into your next stage of cultivation.

Edit: I agree with Rumfor69 on the progress though. You might want to let them set up a little bit more since the colonies are still small.

you're good to transfer whenever you want! i don't know much about transferring a whole colony vs. just transferring the leading edge, but either way you're gonna need to take more transfers so i don't think it matters as far as contams go

I would let them go another couple days then transfer from a leading
edge

I did some transfers.
Not sure what plates are pictured in OP, other than the one labeled "Lite contd" meant to write cond for condensation w/e.

these pics are from today, did these transfers 3-28

Depending on how they look probably use some wedges and either take them to wbs or another plate.
I didn't even take pics of the transfers I'm going to do tomorrow, the green plates on the right are a sneak peek I guess.

Did another transfer today, same recipe. This time I tried to cut time by submersing my agar bottles in 50c or so water, which cooled the bottoms too quickly as It was just hot water in a pot on the counter. They were a little chunky, but only on the very bottom, I still poured them.

The orange dishes are first transfer after streaking from prints, TOC and PESA.
The red dishes are the b+ first transfer.

First 2 pics are updates on growth from b+ transfer.

UPDATE!

Here are the plates today. Some of the b+ have contam, most likely from not wearing gloves in the SAB. :ohwell:
Some look great, they're mostly all about 1-1/4" or larger. The toc and pesa both do not show any contams yet, they're mostly 3/4" in diameter.





Had to delete a lot of pics, sorry.
