Tips for cloning agar pins? Ive been using a small pair of tweezers, need to invest in forceps. Curious if the myc at the base of the mushroom is from multiple sets of ms genetics? Does anyone ever remove the base or only take transfers when myc starts to grow from the center/cap instead of the base? Im probably over thinking it but I want to try removing the base from my next clones with sterilized scissors or something simple.

Your over thinking it. Flame your scalpel. Chop and drop it on a new plate. If it’s from a germ plate most likely you’ll have to clean it up with transfers anyways.

They are all 2-3 transfers from the clones or pins. I like to leave a few plates to age and pop up pins, these usually smash the pasty. Not isolates but some are oins from clones 3 transfers in. Isolation is narrowing those genetics down

i get that growth sometimes and it's usually just mycelium , it could start sectoring after that

Why do people cut such large chunks from their agar? That's not sarcastic, I'm genuinely asking. It seems to me like an empty pointed syringe would be the best tool for transfers. You just knick the surface with it and scoop an almost microscopic amount amount of myc. Keeps contams away and narrows genetics down to a more uniform set. 2 for 1, right? Or am I wrong? I'd love to hear what you think

Transfer a sample of just the outside edge of the clear shiny bit and see what grows. Maybe we can learn something.

Seems to usually be people just getting into agar and they have trouble handling smaller wedges during transfer.
Mine were never very big but as I got more experienced they definitely got smaller.

You can definitely just stick the syringe into one plate and then to another and you'll do a transfer. It just takes way longer for myc to recover and ain't nobody got time for that.
