I just made my first batch of 35 sugars dishes, I am overly excited and have been taking as many samples I possibly. I even got a few samples from a wild oyster mushroom I found, but my question is where do you guys take tissue samples from on yours cubensis for the best results? And how long will it take to see growth? Any other tips would also be greatly appreciated!

For cubensis the inside of the stem is best bet, flesh that hasn't been exposed to open air and not near the cap you don't want any rogue spores on your clone samples.
You'll see it starting to grow out within a few days, lower temps will slow things down.

There's no need to take lots of cuts from the same mushroom, just a couple will be enough you want to keep some plates so you can transfer them n clean it up

As they said, just want to add some points. To be on safe page, you may get multiple cuts, yet do not waste your plates, you can use 3 different area on the same plate. They are all the same genetics so no problem doing that. If one is bad you can continue with others.

Thank you, and good call on the transfers I wasn't even thinking! So two more questions, how do you store your trays? And if the middle of my mushroom is basically hollow do I cut a piece off the inside wall?

Check out Stro's Cleaning and Isolating On Agar. Very useful information in this thread.

Get a scalpel or an x-acto knife and slice a piece of flesh off onto your agar dish.

Here is a thread by FrankHorrigan cloning a yellow oyster mushroom.

Store dishes in a zip-lock bag or on the shelf.
