One good agar plate
    #26566152 - 03/29/20 10:33 PM (3 years, 9 months ago)

Here's my basic circumstance. With a dried specimen i tapped and scrapped spores and gill material onto agar dishes(plastic dishes with green died agar). After the first 3 days they all looked promising. But after 5 days all looked contaminated but 1.

So i have 1 good agar mycelium dish growing at the 6 day mark. I have 5 new agar dishes i can use..... And heres the fun part im sure ill get some love for lol... I have no pressure cooker... Best i can get my hands on is a power pressure cooker xl (7.4psi i think) .... Anyways im in dire need of help. I need to keep this specimen alive. I still have the dried mushroom and cap but not able to get more agar after this for a long while. So i need as many ideas of how to keep this thing going. It can be storage, spawn jars, or some other means. But any ideas or help and as many as i can get is very much appreciated. Ive did basic research online already but i think the community would really help me here. Im new at the hands on part so tips are helpful too.. I can post pics but i already did in a previous thread. Also ill answer any questions if it will help me get somewhere with this. Much thanks ahead of time!

clean it up on agar with a few transfers and put it in the fridge to store it

To prevent contaminating your last 5 Petri dishes;
What is your process for inoculation?
Still air box?

Paul Stamets wrote in a book that he sometimes lets the mycelia run through a glass tube of a sterile mix of substrates, and it may crawl through the tube faster than the contams, being pure on the other side. I'm not sure how to set this up (aseptic technique).

so i basically ended up making a tent with plastic on my kitchen counter. cut a hole in it and used another piece of plastic to cover the hole. and the one that ended up with no contamination was one that i just picked up a mushroom cap with gloves on and tapped it with a flamed metal scrapper tool on the top to let the spores sprinkle onto the agar. two i did by tapping the cap and two i took a flamed scrapper tool to scrape gill material out and made a zig zag pattern onto agar. only one that survived with no contamination (i think) is one that spores were tapped onto the agar. all under this tent. i also sprayed inside of tent with lysol before i started. this is two pic of the one i think has no contamination. i can post pics of the others too. i thought they were good until i held them up to the light and seen that they wasn't translucent like the one in the pic..... im sure i could have been btr off with a sill air box but it was one of those in the middle of the night things. and i didnt really have a clean box without scrubbing it..... thanks for your time. really appreciated.

" it was one of those in the middle of the night things. " that's not the best way to do things, no matter what you are doing

the plate looks usable to me, I'd take transfers from it to the remaining plates
