Props to Ovioid for showing us how to build an SAB that makes you learn good sterile technique, if you move to fast with this one you will slit your wrists.





It worked! I took transfers from these and put them to pasty plates I'm now on T2's I'm gonna transfer one more time and do grain to grain. I might use a T2 if it's clean enough.


My SAB must be working I've had maby 3 of 15 contam with bacteria. Who cares about a few scrapes on your arm it's worth it for sterile technique. :cheers:

I know a lot of guys spray the pc down with cool water to speed it up, I guess it says you can in the manual but it’s pretty heavily debated whether or not it weakens the integrity of the pc over time.

That’s a great way to keep your SAB on the DL. If anyone sees it, they’ll just think your “storage container” took a bad fall down the stairs.

you can usually get a  4" hole saw bit and return it after use;).  they can be spendy

Quick question:

I have some grain jars prepped and ready to knock up. I have some agar plates that are fully colonized (and wrapped) and would like to try putting em to grain to preserve the culture. I know its not ideal with fully colonized plates, but worth a shot? Or would you say dont bother?

I’m guessing its a clone? Do you not have any more plates prepped, because that would be the easiest way to preserve the culture. If you feel some kind of way about it, you can always just stay away from the edges when slicing your wedges. Ha, I’m a poet and didn’t even realize it.

Yeah good call, ill make a plate of it too. Thanks mr Poet ^^. They are clones indeed. Not a super big deal if it goes south, but I may want to return to it in the future. Ill just try it, cut it up to exclude the edges and knock up 2 jars with it and see how it goes.

You can cut a wedge out of the plate but I would stay away from the putter edge of the petri.

I use fully grown plates fairly regularly, but I do leave about 1/3 inch of agar around the edge. Usually just cut a circle all the way around then drop the middle.

I was more concerned with slow growth but hasn’t been an issue, last week I used a plate that was knotting for the first time and it’s actually moving faster than normal.

Ah sweet, good to know. I indeed cut around the edges of my fully grown plates. 2 of them were knotting already as well. Curious to see how it goes. Thanks for your input my dudes.
