Agar syringes

Wondering how your experience has been making syringes from agar plates. Can’t find much info on it here. Assuming just scrape up some mycelium and add it into the sterilized syringe. Shake well. Thoughts? Thanks guys. Trying to keep busy during this “covid lockdown”

I have sterile grain spawn ready. In bags with injection ports. I’m working with a glovebox so using the injection ports with a syringe seems much safer than a grain to grain, or cutting open the spawn bags and dropping in an agar wedge. I’ve seen some teks advise to squirt the sterile solution out if the syringe and into the Petri dish. Swirl the solution around in the dish, and suck back up into the syringe. Then inoculate. Others deposit an agar wedge in the syringe and shake. Help me out guys, I’m not sure either way. Y’all rock!

I have heard of people taking agar wedges and putting them in jars of sterile water and shaking the shit out of them and finally pouring them into grain jars, but never agar syringes

I’m going to shoot some sterile water on the agar plates, stir it up, and suck it back up. Then inoculate with that.

Any more input would be totally awesome. It’s in isolated B+ strain, the first grow with this in ms was surprisingly incredible. So I want to see what the isolate will do.

Drop a wedge to the grain, you’re just adding contamination vectors with anything else.

If you got squeaky clean agar why not make LC and get syringes full of that?

I successfully inoculated rye grain bag with SHIP I injected sterilized syringe with water into a fully colonized BRF jar, shook it up withdrew as much as I could and injected that not the rye bag. It worked with no issues. Took a while to get going though. Probably better off making LC

You may not be able to suck it back up so easily without using a blender.

You can have an LC in an 'LC syringe', but not a syringe with a needle tip, unless you find a great way to chop it up in the liquid- and without a needle tip, you can't purge through a SHIP, although they also make ports for an LC syringe, specifically.

I guess it’s technically an li.
Usually people squirt water into colonized grain jars, shake it up then suck it back into the syringe.
I’ve seen threads with people using plates no problem. As long as any microscopic piece of myc gets into the syringe and then into the grain it’ll do fine.

I thought this was pretty common. A couple times I made LI syringes like this using no pours.

I've put plates in a blendder and then aspirated into syringes. Its one way to do things. Its not my fav way but its a way.
