Spore to agar then agar to grains

I want to know if I can inoculate agar with spore and transfer a few times then inoc grains without losing multiple sub strains. Obviously when you go into isolating your working with very minimal strains. I want plenty of sub strains to fruit so I can then take clones. Just wanted  to know if I'll get multiple  strains even though I'll do a few transfers of multi spore myc

That's exactly how you want to do things. You can make up a couple
multispore dishes to take transfers from, taking transfers from different
places in those MS dishes too and you'll end of with lots of different
genetics to experiment with.

Okay cool. So your better off doing more plates instead if just doing one drop on a  plate? Ideally I want to inoculate 4 to 8 jars with agar from ms to fruit out clones. Should I mix multiple plates when inoculating grains?

Theres a ton of strains in every spore germination. Mixing different agar wedges in the same jar isnt necessary for that.

Isolating one strain is not easy so if you want multiple strains in one jar, its as easy as spores to agar to grain :thumbup:

Nah dude, you can easily germinate on one plate and just do multiple transfers from that one. And when inoculating, just go for one wedge to decrease the amount of contamination vectors.

There's hundreds of genetics on there. You read about non-fruiting monocultures somewhere and I highly recommend you to completely forget about it for a long while. Focus on your SAB technique for now. Contaminations are your main enemy and if you can nail that down, you'll have plenty of time and materials ready to focus on way smaller issues later. And non-fruiting culture is pretty much at the end of that list.

Go read some of the popular threads. People got 99 problems but a non-fruiting culture ain't one.
