Inoculation question

Hey folks. So I'm growing ATL7 screlotia truffles for the first time. I have 36 wide mouth quart jars. I've modified all the lids with poly and red silicone SHIPs.
My question is how often would you flame the needle? I'm worried if I flame after every jar ill end up evaporating half my MS syringe.
Second question, since I have the SHIPs am I alright to do this in open air? My SAB is only 66qts so it doesn't fit many jars at once.

How lucky are you feeling?
Flaming the needle and giving it a little squirt between inoculations isn't going to evap your syringe. You lose only a tiny bit per round.
SAB beats no SAB every time. It's worth the extra time and hassle to use it if you have it.

I flame sterilize between each jar (you don't need to worry about the heat evaporating the spore solution). You could maybe get away with not using a SAB but I wouldn't recommend it (mold spores be EVERYWHERE). If I were you, I'd just fit as many jars in the SAB as you can, inoculate them, take them out, bring more jars, inoculate, etc..

Also, you can just crack the lids inside the SAB and squirt through the opening without punching through the SHIPs to further reduce contam vectors. But you're using a MS syringe anyway, so I'm betting you're a gambling man.

Personally I would cover your lids in foil when you PC, load a reasonable amount of jars into your SAB, flame, pop the foil, inject them all, then reload your air box, flame the needle, and go again until you're done.

I spawned some popcorn casings and had double-overlay cause I didn't put enough hydrogen peroxide in my automated aquarium mister. I only got one mushroom so I cut off the head part where the seeds fall from and put it in a jar of LC and sprayed it all over a tin of PF cakes I made with gravel, cardboard, and bisquick in my microwave. I think it will be good cause B+ is so potent. Triggered yet? Only a square would say "a cube is a cube."
No, this does not look right...

Yeah, even as I was typing the question I started forming this plan. Flame once for about 8 jars, swap'm out and do it again.
Although I don't use foil anymore. My poly is always wet under the foil regardless of how long I let it sit. Whereas it is always bone dry when no foil is used.

Punching through the SHIPs is way less risk than cracking lids. It's literally their function, that's why they're called Inoculation Ports.
And MS is not really a gamble when you're growing truffles, that's more applicable to fruits.

Fair enough. Everyone has their own opinion on SHIPs, but as long as you're working cleanly you'll be fine in either case. If a MS truffle syringe is more resistant to contams that's a definite bonus. Hope you get some good stones!

Using those silicone ports get old quick. I spent a bunch of time making them in my lids and ended up just cracking lids in the SAB. The idea is nice, like fixing gloves to your SAB holes to fully “close it off” But in practice it doesn’t hold up.
And using an MS syringe is always a gamble because you just don’t know how clean it is. Random genetics is one thing but unwanted spores can definitely be found in reputable syringes.
I hope it works out for you though. Growing stones is cool

Yeah, it's possible to get a dirty syringe but I've never gotten one from SW. And I've got 4 syringes so even if one is dirty at least all my eggs aren't in one basket. Especially important with truffles which take so long. I'm excited to test for myself the potency differences between a 3 month truffle and 5 or 6 month truffle.

Glad to hear you've had good luck with syringes, but it's impossible to have clean spores, syringes or prints. It's just a matter of how dirty they are. This is why people bother with agar.

Man, I have not been on this site in 6 years. What is a SAB?
Also, always flame the needle until it glows red. While it is STILL glowing, inject a squirt of your culture into the mason jar and then immediately cover-up that whole with medical tape.
Get a 1/4 pint mason jar. Fill it halfway up with rubbing alcohol, put the lid on, make a slit in the top about the length of a quarter. Make sure the lid is tight on the jar...then thread get a cotton ball...stretch it out and into what looks like a wick. Then thread the cotton ball through that mason jar lid and place it just deep enough that the bottom is saturated with the rubbing alcohol. There you get a little makeshift bunsen burner.
