5 years old golden teacher and mexican spore struggle

Hey all
After 5 years, I started to vaccinate again. however, the sports remain unresponsive. I have been vaccinated with syringes on the patato dextroz agar and rye behind the necessary sterilization and I am waiting. there is movement in some jars, but it is not clear what it is.

That first one looks like it might have some decent myc at the very top maybe you could pluck a grain.
I think you need agar or if not that brf.

I will take the sample from the rye and put it on the agar. I will try to get fruit from the jar (for fresh sports)

there is a smell in the colonized jar. I put one rye grain in agar. it grew and I cloned into another agar. Since my spores are very old, I put most of my spores in pure water with honey.I took an example of LC from 5 years ago and put it in the agar . spores that I kept in pure water for 5 days still have no effect on agar and keep waiting.

I've hydrated old spores (really old ones) in distilled water with success at germination. It can take weeks or months though.

I'm so confused by a lot of what I'm seeing here:
#1 why are you wrapping tinfoil around the sides of your jars?
#2 if you are getting contams on grains, you will definitely get contams on an lc using the same inoculant. Its never a good idea to put spores in lc, because spores are never sterile
#3 what kind of lc prep are you using for those bottles?
#4 I dont think you are using that hydrator correctly. Putting an airtight sealed container (like spore syringes) inside pressurized container probably isnt going to actually pressurize the liquid inside the syringes. You could just put a rubber band on the syringes instead

Dear seratonin, 1-) I use the foils as partially breathable cover 2-) I used more than 10 different prints, all 5 years old.
I have to take the risk of LC contamination. Do you have a different method to germinate older sports?
3-)a-) pure water + golden teacher spore b-) pure water + spore+ gentamisin c-) pure water + O2+ spore D-) pure water + dextroz + spore
4-)The sample container in the chamber has ventilation. If you have a chance to try it, you can observe from the lines that the syringes react when the pressure rises.

It just takes time more than anything else. For a 15 year old print of Ps cyanescens (stored under constant refrigeration) straight to agar I got germination after 3 weeks. Cubes are known to be less durable so you might have to wait a long time. I got germination from a 6 year old swab of PE after a month so judge appropriately.

thank you, your message has been promising for me. I hope it will germinate in my spores. because I live in a restricted country.

I bought a jar that I suspect was a golden teacher in the fruit room If I can reach a fruit, I will be very happy and I can bring more

Op I bumped your other thread.
I'm having trouble with the hydrator thing you built and would love to see it work. It just seems that water is incredibly hard to compress and their wouldn't be enough force in that jar to push the plunger more than the rest of the syringe.
So I guess what I'm saying is that if you are increasing the air pressure in the jar at even rates your applying as much force to the sides of the syringe as you are to the back and tip. So does this work, or do you need like glass syringes and an air tank with a compressor and regulater hooked up to it ?
Cool gadget though.
