Agar xfers

Full disclosure, I suck at agar transfers. I have a hell of a time getting the wedge off the scalpel without smearing the wedge all over the plate like butter on toast.

These cultures are growing on pda purchased online with an average room temperature of 64 degrees Fahrenheit.

I have uploaded pictures of the front and back of the plates.



what looks good?

what looks bad?

Are any of these ready to be dropped in grain or should I transfer again?

I have the materials on hand to make lme if necessary. I do not have the experience, this would be a first time experience for me.

None of that is anywhere near ready to go to grain.

But you have plenty to work with, keep taking transfers.

I'd avoid the ones with yellow, and the ones with tiny spots you need to let grow a little more.  The others are ready for transfer.

I also use to suck with transfer for that exact same reason, the game changer for me was a #7 scalpel and #15c. I do like 30-60 transfers at a time now.

try using a softer agar. I had issues too with harder agar cutting with a scalpel.

I made some lme agar dishes today. Mistakes were made in the process. I used 500ml of water instead of 400. Then I sterilized the mixture at 18 psi for 2 hours. There was some caramelized chunks floating inside, I think the extra 100ml of water saved it from becoming chunky soap.I poured the plates anyways hopefully they work.

I transferred again, and made a mess again. I will look for #7 blades and give it a try. My technique is probably bad.

#7 is the handle, the blades are the 15c

lol I think its sediment or caramelized agar.
I get this too sometimes when i do BRF agar

Do you think the #5 could be put on to grain? Or should I wait till the others grow out? I have a bunch of oat jars in the fridge waiting to be pressure cooked.I got a bit ahead of myself.

put it to grain! I would use scalpel to cut completely around the the circle of myc and drop it in grains or a LI.

Lmao, I skimmed bods agar tek and skipped the part where it's only supposed to be in the pressure cooker @ 15 psi for 20mins. I left the mix in the pressure cooker for 2 hours @ 19 psi. Now I know why we don't do that, tadpoles.

Sounds good. I am excited too as I have done about 4 transfers with some RW's. Swab to agar x 4. Now I have 8 wbs jars about 35% colonized using LI. I took a agar plate about the size of yours, then cut it out in a circle and dropped it in sterilized water, shook the fuck out of it and poured into wbs jars. I also took 4 jars and took a plate your size and cut it completely out, leaving the agar and completely circling the myc and dropped into wbs jars. I gotta say, the LI is much faster, but I did both in case the LI was fucked

I'm not fluent on the acronyms yet but I did a search on here. Is RW RustyWhyte and wbs wild bird seed?

How is your progress going?

I did as you suggested and cut a circle around the myc, I then cut it into a bunch of squares and dropped it into one of my oat jars and gave it a good shake. I will take a picture of it tomorrow and post it here and see what you guys think. If it's good, I'm going to go ahead and do a grain to grain transfer and get the rest of the jars going. On the bright side, all of the jars I made last week are contamination free I do not see any signs of growth.

Update on my progress, do you see any contamination?

Does the jar look good to do grain to grain transfers to the rest of my jars?

I have been doing everything using a SAB so far. I have been building a laminar flow cabinet and it will be ready this weekend. However I have never used it so I do not know if it will work properly. Since this is my first grow, should I continue with the SAB to finish this first grow off first, and then test the flow cabinet with the next one?

That spawn is sketchy. Spawn it to bulk and hope for the best.

You'll take better pics of plates on a darker surface.

Okay, flow cabinet is complete!


I went ahead and took better pictures now that I have better lighting:


On the left is one of 10 jars filled with oats. I inoculated this jar with cut up pieces of a single agar dish about two weeks ago. On the right is one of the other 9 jars that have just been sitting on a shelf the last two weeks. I took pictures of each angle of the jars side by side to see if you or anyone can point out to me what looks contaminated and what looks good. I'm trying to learn how to spot contamination and healthy growth. My house typically sits right at 64 degrees fahrenheit 

I had intended to take grains from the colonized jar and inoculate the rest of the 9 jars. Is this advisable? How long can pressure cooked grains stay in the jars without being inoculated before going bad? It would be a shame to let these jars go to waste.

To me the jar looks a little bit bacterial, but I would still spawn. The grain pressed against the jar and the yellowing in these areas is the biggest indicator in my experience.

It's also seen in the uncolonized jar on the right. In my experience, this happened when I overhydrated the grains or didn't let them dry long enough before putting them in the PC. Everyone has a different environment, so it just takes some adjusting.

Personally, I would spawn the jar on the left to coir and take some clones of the fruits that you get. The 9 leftover grain jars I would scrap and try again. It sucks to lose the grain, but it is a good learning experience.
