Agar Overload!!

It crazy how simply follow a tek to the t can bring results. I have struggle with pouring my own agar and had resorted to buying pre pour dishes. Until I found C100 post( Have no idea how to hyper link post if some one would share) I pour 40 plates and success all of them have transfers on them. I was expecting this now I have 50 plates of agar although I am doing 18 tubs. Currently I have decided to use grain slurries to knock up my bags cause it seems to give the most bang for the buck doing all 18 bags with 1 jar meaning 1 plate of agar I think. So I’m wondering with all these extra plates there’s got be some other skills I can practice creating LC, transferring to create monocultures, or introducing new strains. Is there any other teks that begin with agar that I should develop? I’m going to attempt to make my first batch of LC can anyone highlight the benefits of LC? How long does it store, how much can it knock up, and tek reccomendations? I understand how to get to monoculture but have a problem with sectoring for best transfers I’ll include some pictures? Does a monoculture help isolate specific characteristics such as yield, colonization speed, fruit characteristics? One final note let me know your experience with inculcation methods I’m stuck between LI, Slurries, LC for large volume of spawn bags? Best agar storing practices inoculated and fresh pour plates? Look forward to the insight.

Finished 20 transfers today going do another 40 in about 2 days. Got Ecuador, Puerto Rican, Costa Rican, and Golden teacher strains going. Going attempt to knock up 18 grains bags with a slurry tomorrow from one of those colonized jars. Putting them in Patsywhyte ez-dialed in tub with a 1:2 ratio, looking for direction on above questions feel like LC our because hidden contamination. Anyone with experience with slurries?

I only have one grow under my belt (currently still harvesting) so take everything I say with a grain of salt. I have found bodhisatta's TEKS (he has a sticky with a list of teks in his profile) to cover pretty much everything I need. I did spores to agar and then inoculated half my grain jars with agar wedges and half with LC. I tested the LC on agar before using it to inoculate, and being my first LC the results were amazing. I got no contamination and I was able to inoculate a lot of jars and could avoid G2G, which in my small SAB would've been problematic. The speed of colonization is very nice and with 200 ml of LC you can inoculate a ton of jars. I didn't really measure it out since I had excess LC for the jars I had, but 1-2 mL should be more than enough for each jar. On my next grow I will not bother with agar inoculation, LC is just so efficient for bulk that I can only recommend it. I have never heard of grain slurries so I can't really give my opinion on that. Of course you want your sterile technique to be on point if you're gonna do LC as you'll have no way to "fix" any contaminants that might get in.
About monocultures, from what I've read isolating sectors and working with strains is a long process that requires you to test all the different isolates and grow each one out to determine which has the phenotype you like. It's something I will never do as I don't really have that much time and space.
As far as storing goes, I think slants are the most efficient way to store mycelium long term and spore prints for spores. The search function will help you, as well as all the TEKs I told you about above. I'd post a link but I'm from my phone. As far as storing poured plates for future use, I always pour them when I need them. But then again, I have glass Petris, so I never work with too many
