First grow, did drops from syringe onto agar. I now know I should have done at least one A2A transfer, but I had already gone A2G when I learned this. Also aware that I put way too much spore solution onto the plate. Regardless, how does this growth look (is it myc or contams) and what are the chances I will get a colonized jar assuming I did not contam in the inoculation process?
I had about 10 petris that all looked very similar to this one. In the future I will isolate at least somewhat using A2A.

hey bro. It looks all right so far.
I recommend avoiding anything from the edges as it easy to contaminate there (You already have some edge colonies at the top of the photo i would question, Does that growth look like the rest in person?, is it glossy?. is it a contamination?, why find out when you can just leave it behind by doing another transfer?). i recommend doing a2a on a few dishes, then let it grow out 3/4 the dish, then transfer the growth you prefer. It grows a little funny at first so a couple transfers really helps identify it before you waste all your time. Colors may show up in about a week or two. Any Color at this stage would mean a contamination. Green pink yellow etc.
If you put that in a jar right now you have wasted your time doing the agar in my opinion (i'm not all knowing and please don't take offence just my opinion as it does look good at this stage but it changes how it grows a lot, I'm sure someone else will give you there opinion). Lots of contamination's don't show there colors yet and you cant see if its growing how you like (organized or like the strain your trying to grow.) wouldn't want to grow cotton or trich by accident.
Next time if you really don't want to do heaps of transfers do a lot of dishes with a single small dot in the middle. By the time its 3/4 gown out at least a couple will show signs of the right type of growth, then take those wedges out.
I'm happy to see your taking the time to use agar. It show's your on the right track May as well go a touch further now so you can identify your growth better. Anyways...
Good Luck

Hello,
I totally agree about going agar to agar a couple times. I still think that one agar transfer would be cleaner than a straight spore syringe into a grain jar, but I could be wrong. I will try to find some good colonies and transfer them to a new plate.
I felt like the growth was very slow on these plates, they were inoculated with a syringe about a month ago. My spore syringe was missing a needle tip which meant that I could not dispense small drops or flame sterilize it. I looking into buying a needle tip but they were only sold in bulk and I did not need 50 of them. Hopefully that was the last time I will use the syringe.
Thanks for the detailed responce.
Time to pour some more plates, Ape2
