My first agar plates

I recently started playing with agar, here's my results:

T1-10 were poured on 4th of february. The sleeve they came in was punctured so I didn't expect to see any of them survive but 3 of them look ok to me (T1,3,6)



T11-20 were poured on 11th of february. I believe that the agar used for these was too solid and that might be the reason why nothing grew on them at all. I'm not sure if I should keep them or not.



F1-10 were poured and inoculated on 24th of february. I was very happy with this agar because it was a lot more clear but it was still a bit too solid. One of the plates I threw before taking the pic because it looked so fucking disturbing that I literally thought whatever grew on that thing was going to grow limbs and kill me in my sleep.



Three of those F plates look like they have a bit of mycelium growth going on (F5,9,10) so I took 2 close-ups, one with backlight, the other with flash. I would love to know if these look good to you.

Yes, looks ok to me. Now take more transfers, take those 3 plates and hopefully make them into 10 plates with mycelium.
How did you inoculate  your plates? What strength agar recipe did you use?

From what I can see, they're all contaminated but keep transferring.

I used a homemade loop and the print i used was more like a big pile of spores in a piece of aluminum foil. I couldn't properly break up the clumps of spores so that's why there's lots of black spots on the F ones. Also, the agar didn't quite fully solidify when i inoculated so some of the spore clumps got stuck inside the agar

when i first tried 2% agar it seemed to be nowhere near solidifying, so i added a bit more, up to about 3-4, but i think i used too much


The T ones have a huge amount of tiny potato bits in them and the agar was slightly bubbly when i poured them , that's why they look so cloudy and dark


I'll transfer those three and I'll pour some more plates soon

They’re all bad besides f4,9,and 10. Transfer from those, the rest are garbage. The pics are terribly lit as well, so I could be wrong but almost all of them look like you should throw them out.

Yea, sorry for bad lighting, it's my first time taking agar pics
I wanted to take transfers 2 days ago but I didn't have the time to do it so I'm gonna do it today

Here's how those three are looking now

I want to transfer from each plate to 4 more plates and 2 or 3 jars of grain

F3 and 4 grew quite a bit as well but they have contam in them. I was thinking about trying to transfer from these 2 as well when I pour some more plates just to see if i can clean them up.

I don't see the point in waiting more before transfering to grain, perhaps I am missing something?

My plan is to fruit as many different mycs as soon as possible and clone from the best of those. I already took the transfers but if there is a reason not to do it this soon I would love to hear it

I'm currently using a makeshift SAB that is very uncomfortable to work with, but I will make a proper one soon

I think I understand quite well the principles of how how a sab works and I try to follow all basic rules like never hold something that is not sterile above something sterile, but I don't have much experience so I can only consider my sterile work to be decent at best

Definitely do more transfers. There’s all kinds of bad shit on those plates you don’t want in your grain. You should be able to clean up your cultures with more transfers. Most people go with at least 3 transfers from spores, I’ve gone with two before but from clean plates and those aren’t clean plates.

You talk about wasting time doing more transfers, well look at this way, you’ll have wasted all your time and ended up with nothing when your jars goto shit if you use those plates to inoculate and wasted all that grain.  :2cents:

Here is a good thread of what you’re after and what you’re not. It’s up to you if you take our advice or not.

Edit.

Read up on sterile tek, show us pictures of your sab and your tools. I take it you’re flaming your scalpel between transfers? Use the search function there’s plenty of info out there :thumbup:

I agree with your plan, when you are sure you have a clean culture. While I don't see any obvious contams on #10, it's highly disorganized and could be contaminated. I'd continue taking transfers until you are certain you are clean, which is the point of agar.

But there's no harm in doing both. Since you already took transfers, take what you feel is a healthy wedge to a jar of grain. If you can grow it out, you'll have your clones. If it contaminates before fruits, keep cleaning the agar culture.

That's pretty much what my thought process was

I wouldn't attempt trying to transfer to grain if i wasn't pretty confident that those plates are clean but I see why you say they look dirty. I think I'm just really bad with taking photos. when i look at the pic there's some parts that def look like contam but I'm pretty sure it's just the light reflecting in a way that makes the myc look yellowish, smooth and kind of glossy, I might be wrong tho.

The grain jars I used were only about half quart in size so there's not much material/time waste if they all go bad.

I'll update in about a week when the transfer plates grow a bit, with proper photos this time (hopefully)
