First attempt at agar work - where to go from here?

So, having just getting back into this 30 days ago, and starting my 2nd grow with a GT syringe bought from a reputable company (promoted on this site), I put a drop on a couple different agar plates, got some good mycelial growth, and took a piece from the periphery of each plate and transferred to 2 more plates. My question is this...how many transfers to do, and what am I looking for to know when I'm "there". If you look at the pics, the T2 plates are showing some "sector-less" growth on the right side - is that what I want? I'm really digging doing my own agar, but need some educating on the finer technical points. I also cloned one of the better fruits from this grow, and am now dunking after my 1st flush. Your input, as always, is much appreciated. Oh, btw...I really want to get into doing some Semperviva mycelium for a microdising stack with Lion's Mane and Niacin, and wondered how to go about getting spores, as the online resources seem to not carry it, or they are out of stock.

When inoculating from spores, there will be numerous strains on the Petri dish. The different growth pattern you referred to as (sector-less) is actually sectored, representing multiple strains on the plate. There are a couple different types of growth patterns like tomentose and rhizomorphic that can be expressed. Essentially you are looking for clean mycelium free of contaminates. However many transfers that may take.

I would do just a few if your not getting contams. I don't know how to identify the best parts to take, but I look for little spokes and uniformity. You could always do few transfers, which you have done already, and transfer to grain. At least you will have clean spawn. Then clone from tissue.
Josex biopsy method seems kinda kool too because you can do a poke and hopefully narrow down a strain and do few transfers for cleanliness test and if all is good, use that for grain or spread to more plates
