So I order 500 Petri dishes and forgot all the different sizes will these 60mm work, I read on different forum that clones in these performed better? What’s your thoughts?

They are fine you just have to transfer before it reaches the edge which will be much quicker since you have smaller plates

I only let them grow so much before making transfers anyway , the 90 and 100's I used to use when making isolates

I like 100mm. I'm paying $65 for 500. What do the 60mm go for?

Additional question concerning agar, I just pour 40 plates last night and was wondering how to do the control plate so I don’t do 40 transfers on to dirty plates?

I use 60s. Makes the agar go alot further even if you pour thick.
For a control plate just leave one without innoculatimg and see if anything grows on it.
Or just wait a week or two before using and check the plates.
If I ever get a bad plate from pouring it will be like 1 out of 60 plates. So unless you messed up pretty bad they should all be good for the most part. If the agar and blanks were both sterile you will be ok.

I like the 60s, especially if you're doing a ton of transfers just for the fact that you can stretch your agar. It's been a while since I've poured 60s but I feel like I could get 40+ dishes out of a 500ml bottle.
But think about when you're doing a bunch of transfers, you're gonna take your transfer well before the myc gets anywhere close to the edge of a 90-100mm plate.
I also used them to drop wedges or make LI no prob. 100mm plates may be better for that purpose but your 60s will certainly get use.

Great info looks like I got 40 transfer today, going pour another 40 tonight start some fresh off a MSS. What’s your guys best practices for storage of unused agar plates I’ve seen ziplock to just stick them in the fridge? Should I parafilm them?

I just let them cool completely then put them back into the sleeve they came out of. Then I told the edge of the sleeve over and put a piece of tape on it. I used to put them in ziplock bags but that got old fast. Bags get expensive if you use enough of them. You can parafilm them now but that's alot of extra work putting it on taking it off to use them then taping them again. I wouldn't store them in the fridge. Just in the open or in a tote or something.

I'd prefer 60's over 90's or 100's. Things get slick with iso soaked gloves. The 60's would probably be easier to grip.

I've used both 100's and 60's. They made no difference to me. I'd go with whatever you can find for cheapest.

I was wrong. I had 500 for $65 saved in my favorites and I thought that's where I bought the batch. But I just went to order some more and the shipping rate was absurd. Found them 500 for $75 plus tax with free shipping on eBay from Fablab.

That makes sense. 60s would be more economical for transfers and 100s are better for taking wedges.

I use the 60s in 10 sleeve bag. I find it easier to work with them in my sab.

Same. I like sleeves of 10. 20s and 25s fit but not comfortably.
