is this agar plate contamed?
    #26514906 - 03/03/20 04:16 PM (3 years, 11 months ago)

I made some agar dish ready and waiting for my older plates to grow and do the transfer . But this is now on all my new plates
This picture is taken from the bottom of the plate
Can this be because i used tap water?
Can this be because i reheated an stored agar(for one day in fridge)
I initially made 4 petri and everything went perfect , now i did 16 and all of them look like this

Tapwater is fine. Storing the medium in a fridge for one day is also fine.

Describe how you made the plates.

well like the first time i used my gloves and mask
i used alcohol to clean the air and surface
i turned the flow hood on
putting everything i need (jar of agar - petri dishes in plastic) in front of it
i opened the jar and poured some in each plate, now since this was my second time i messed up a little bit like while pouring but this should not affect all the plates!
and i closed them with selefon and stored them at 25c
been like 1-2days and i noticed these particles , i dont know if they were there the whole time or just appeared

edit : btw the recipe was 10g agar 10g malt extrarct 500ml water
pressure cooked for 25minutes

Hmm, i pressure cook agar bottles for 45 minutes to be safe, but people do 20 minutes without any problems... Did you vent the pressure cooker? Also, were the plates elevated while you poured them?

i let it release by itself since the last time (even tho it was successful it spill like half of it)
plates were in my hand and elevated

now that i think about it i can think of one other thing that could cause this
1-i used an old grain jar of mine which halfway through stalled - but i cleaned it with soup and stuff and since this can be bacteria shouldnt pressure cooking it kill it?

what i meant by "vent" is leaving the valve open for like ten minutes before closing it and beginning the pc cycle. This releases the air from the pressure cooker and allows it to fill it with steam. If you don't do this, the pressure cooker will not reach the desired temperatures.

i guess i didnt do that
should i wait to see if these particles grow or not?
if they dont grow at all for like a week , is it safe to do the transfer?

im not sure if i would risk it, but if you have nothing important to lose than it would probably be worth the try. I think that not venting (in other words - insufficient sterilization) was your issue, since the contam (if it indeed is contam) seems too spread out for it to have gotten into the plate while it was open.
Venting is important. I did not do it when i was pressure cooking my first grain jars and have gotten horrible yeast contamination. I started venting and have been yeast-free since then.
:goodluck:
