Clones from growkit - weak mycelium?
    #26507748 - 02/28/20 07:46 AM (3 years, 11 months ago)

Dear Shroomerites,

Just a general question for you:

Is it possible that taking clones from a growkit yields weak mycelium, no matter which shroom I clone from the same growkit?

I dont fully understand if its senescence (that even possible when cloning a shroom?), or its my grains/sterile technique. Fact is that my cloned myc grows very slow on grains and recovers slowly after shaking jars. I took multiple clone samples from different flushes of the kit, with similar results. So I was wondering if the kit is maybe already old mycelium (many transfers in from original spores) which results in weak clone tissue. The kit itself yielded above expectation though.

What do you think? Is it a possibility or am I fully to blame here

Any opinions on this? Or maybe an existing thread that explains the science behind clones/senescence? Been reading a lot but find it difficult to find an answer to this. :takingnotes:

^^^ senescence discussion thread.  Not fully understood, even by Bod, Workman, etc... When you say "taking clones" do you mean taking to agar and cleaning through transfers?  If you are experiencing slow growth and stalling in jars you can generally thank mycelial growth encountering contamination (slow/no recovery post-shaking is a good sign of this).  You should get perfectly good clones from the fruits from your kit (no reason to worry about senescence in your case, so best to refocus on your technique and go through all the steps you used to find the hole in your sterile work.  Take us through what steps you used to obtain your clones, grain prep, sterile work, etc and we'll be better able to assist you.  Some varieties/gentics do run a little slower, and some run a bit faster than "average".  Again- more info needed.

Thanks for your answer! Thought this was the case.

Im indeed cloning from shrooms to agar, using SAB and no visible (to me) contam on the plates. Suspecting its my grain prep as my newer jars which are more wet do seem to grow a bit quicker, as are my jars with different type of grain. Too early to tell though still, was just curious if it would be possible to get weaker myc from myc that has been expanded for a while, which probably is the case when buying growkits. Ill focus on getting a good moisture balance in my grains. I guess it takes some trial and error starting up getting the balance right.

Good plan.  Too-wet jars take longer and promote bacterial growth (although that bacterial growth is only casued by hitchhikers in your spawn or inadequate PC time/pressure.  It can be really hard to spot hidden contams on agar..  "One more transfer" is always a good plan, as is posting on an agar thread here to get a second opinion if unsure.  If you do those three things (agar work, SAB tech, and grain prep), you'll have eliminated variables.  Best of luck!
