Not sure if I injected enough spore solution...
    #26507909 - 02/28/20 10:08 AM (3 years, 10 months ago)

Hello everyone.  This is my first time logging bon in a long time.  I have some rye berry bags I just tried to inoculate a few days ago.  I realize I may have mad a few mistakes now looking back on it.  I purchased some syringes online.  They seemed really thin (especially compared to what I normally make), but I didn't worry about it because I've always heard about how even if you don't see much in the syringe, there is still lots of spores.  Well, I ended up injecting a few CC's of solution per 3 pound bag.  I made the mistake of shaking the bag right after injection.  Thus far I see one tiny speck of white in 1/8 bags.  I wonder if me not putting in enough solution, AND me shaking the bag may have messed things up.  I'm hesitant to taking another syringe and sticking each bag a second time, because I really don't want to contaminate the cultures.  I realize this is a patience type of game, so I'm probably just going to let then go as is.  What is your take on this?  Should I put more solution in at a risk of contamination, or should I just let them go?  I've heard that at least two spores have to be in contact for it to take off.  Thank you very much for your consideration, and your time.  I hope you all are having a great day.

Chances are pretty good that you already contaminated your grain. You should never inject grains with spores because spore syringes are always contaminated. It can work out, but in the vast majority of cases, it doesn't.

Yeah, shaking it was a bad move, but you will probably see some growth eventually.

If i was in your place, i would try some easy no-pour agar tek to get a clean culture... In the meantime, you could do some brf cakes... The success rate of knocking them up with spores is higher than with grain. It depends on how contaminated your syringe is.

I never had a problem starting spores on wild bird seed, but I also never had weak syringes. I usually put a whole print in a syringe

I appreciate the feedback.  Yeah, I figured agar would work better.  I don't have any fruits on hand at the moment, I put a few drops of spore solution on a few dishes last week.  I used some of my syringes, and bought some. I did that to get some good agar, but also to see if my syringes we're contaminated because I've had problems lately with them.  So I ordered some as well, and wow, the difference between my syringes and the store bought is crazy.  I never realized how much stronger I made mine up.  I didn't know there was a way to put inoculated agar into rye bags.  I thought that was just a jar thing, and you had to use LC on bags.  Thank you for any an all information!

Do either of you two have any ideas for a good LC tech that I can look up?  I've tried honey, honey & Karo, and they have all failed.  Not sure what I was doing wrong. Bim going to try it again later today I feel.  Thank you all for the information.

You could try my Maple syrup LC, here is the link:

For me, it worked much better than honey lc.

I think agar is your best bet.

I'll have to look into hot to use these agar dishes I have going right now in a presealed grain bag.  I'm sure it's out there, but I thought to use agar, one had to use jars.

The amount of spores in your syringe has no bearing on your rate of success, regardless of what you use for spawn.  A whole print is super-overkill for a single syringe.  Less is more, and using more spores is actually upping your chances of contamination (despite any random success stories people report; as they often do taking spores to grain).  Speaking of which OP- I'm guessing you ordered grain bvags online as well?  Not at all trying to bust your stones, but online grain bags are a real hit and miss, often ending in bacteria-induced sadness.  Just let them go and see what they do over the next week or two.  Go grab some agar at the asian market or Wally World or online and save yourself the headache.  If you have any spore solution left you have more than enough to start movinng forward with agar.

Some light reading while you wait for your agar.  All you need to know.  Best of luck.

I used a BRF cake that was fully colonized injected sterile water and syringe into the cake before it was birthed shook it up then after flame sterilized the needle withdrew the water and injected that into a pre sterilized rye berries bag. It took a while to colonized , not as fast as LC. I used the bag for a shoebox with striaght coir and got a good harvest
