Bacillus?
    #26503386 - 02/25/20 03:16 PM (3 years, 11 months ago)

These are bacterial, right? Is it Bacillus? Can a jar be assumed bacterial as soon as the grains get this "red" tint?
:thanx:

Its hard to say at this point, but I'm leading towards yes. I don't see the red tent you're referring to though. Also no one will be able to ID the type of bacteria without a microscope and multiple stains.

Most likely you are prepping your grains too wet, you will be able to tell if you get wet spots after PC cycle, before inoculation, wet grains will make it difficult to colonize and promote bacterial growth

Maybe not red, but this orange-ish/light brownish unnatural tint. It is more visible in this photo.

The jar on the left is pretty much how all my grain looks like after pc. This one was pcd three days ago.

I think that there was hidden bacteria in the donor jar and therefore it showed in all receiving jars...
Would you guys toss it, keep it, or wait and see?

I would wait and see. But your grains look good after PC. Probably an introduction of bacteria during inoculation if anything, I see some yellowing metabolites it looks like in the right hand picture, which is usually a sign of infection. If your grains are looking like that before inoculation, definitely seems you are introducing a bacteria during inoculation, your starting material is most likely contammed, syringe, or agar?

Also not sure if that paper towel over top is your filter to open holes, but that could very likely be the cause of your issue as well

my starting material was agar (third transfer from clone tissue), but i took a biopsy from the plate, injected it into lc, let it grow out and then used it to knock up jars...

On the fifth transfer of that same culture, some bacteria showed, but it was not obvious how it was introduced into the plate. I think that now i know.

Try using the previous plates or onces that fully grown out and are free of bacteria, can be tricky to spot sometimes

yeah, i think that in the future i will just make parallel agar cultures and use them to knock up grain - then g2g.
