Anybody with the 25x, I’ve got a question about PC’ing the agar media bottles.  Are you putting them in the insert container filled to the same level or are you putting them on the rack over the heating element and if so, what are you doing to keep exhaust tube out of the water?

I tied my exaust tube in a knot so it doesnt dangle.  I like just on the rack  but I put another rack on it to elevate it above the water level.  the insert fills with water and I dont like that on the bottom of the container when I remove it

I’ll have to do that, dunno why that didn’t cross my mind....thanks for the tips

I didn't remove anything from the interior just cut off and discarded the end of the wires where they were burnt and put new ends on them and just reconnected it. I'm a little afraid if I would ever have to replace the insides as i'm not that apt with electrical. sorry I can't be more help with that.

ah, gotcha.  the wiring aspect is pretty simple but the reconfiguring of said parts...:facepalm3:  I know where they go but how the hell do I make em fit again without breaking... I dunno.  after the 1st time my temp knob stuck out an extra 1/2 inch or so and was crooked. worked fine but looked horribly off.  now its all melted again and I wanna fix it proper

Saw you guys talking about the 7 handle and thought it looked cool so I ordered one up.

So much nicer than the 3 I’ve been using. It also made me realize my first handle wasn’t made right and the blade wasn’t sliding all the way on.

I picked up a box of 100 for $6.99 free shipping.

Individually wrapped. The bitch is trying to get it on the handle so I ditched it and used my xacto knife handle instead. I just keep the handle in ISO and use a new blade for every transfer.

Are you sure you were doing it right? They just slide right onto the handle you
definitely shouldn't have to force it in anyway

Agreed! Should be easy and not have to force it. I tried for like 15min and the tip of the handle looked too tight for even a piece of paper to slide into.

Here’s what I do to work as sterile as possible:

I open the sterile blade packaging just enough to expose the end that slides onto my xacto handle

I do not expose the blade until I’m ready to slice.

I set it across the little dish of iso the xacto blade handle sits in after I’ve cut and removed the blade

I remove the foil from the plates I pc’d (pasty plates tek), wipe the lid down with iso soaked paper towel. Carefully lift the edge of the lid to break the seal but don’t remove. I grip the lid with my fingers to loosen the entire lid off but don’t actually remove it.

I remove the cling wrap from petri and wipe the outside edges with iso soaked paper towel

Now I take my blade and find the thickest myc, avoiding the edges and cut a rice size piece

I carefully slide my hand over to the right, put the lid back on the petri with the other hand, then take that hand and slide the lid 1/2 off and dip the blade with culture into clean agar

Never do my gloved fingers touch the inside or even go over the new plate

Does this happen to anyone?

I struggle in getting the little piece of agar off my blade. I find myself smacking the handle to the edge of the plate and am worried I end up knocking contamination into the new plate!

All your feedback is greatly appreciated. I’m a total noob and after p tonight p, I’ve made 65 transfers.

Foil on your plates? You don't need/want foil wrapped around your petri dishes.

I like #15c scalpel blades and #7 scalpel handles since they make it much easier to avoid this problem.

Plus my bacticinerator sterilizes the long neck of the #7 scalpel handle as well (without burning my hand), which also reduces the chances of contams falling off the handle if I did want to bump it like that

Its badass, it's a standard bacticinerator. Just plugs into ac outlet.

Hands free scalpel sterilization:) takes about 7 seconds to get red hot

Lol exactly, and it frees up both hands for labeling! I used to flame with one hand and label with the other

Also way safer than an open flame when dealing with spray bottles and rags full of alcohol.  I fuckin love that thing

A little bit lol.. my handwriting is equally horrible with either hand

I got mine for a stupidly low price. 37$!! Theres an identical one on Ebay now for 100. I'd happily pay 100 knowing how handy it is, if something happened to mine

The foil around the blade it comes wrapped in. I peel back just enough to expose the end that slides into the handle, slide into my xacto and don't take off the rest of the foil wrapper until I'm ready to slice into the agar to minimize the air exposure to the blade.

Once I take the cling wrap off the petri, then I remove the blade wrapper and slice, transfer and discard the blade. One blade per petri.
