Hello,
So made a sporesyringe and inoculeted both agar plates and liquid culture (LMEA).
The problem is theres is no growth or extremly slow that the bacteria can take over.
Im wondering if the spores i used are to old and somehow dormant.

For LC,i used 0.667g LMEA to 400ml water and PC 30min.
For agar also used 2% agar and 2% LMEA.

What do you guys think is the problem?

The problem is that you put spores to LC guaranteed failure.
But yes besides that spores can become unviable after some time.
I missed the part where you said how old the spores are.
If they have been stored right 4-5 years is not unreasonable.
Hope you have some luck. :thumbup:
Spores can easily take up to two weeks to germinate especially older spores.

I've had better luck taking old spores straight to agar and skipping the whole syringe step. How old was your print?

Location, location, location.

All of my 10 year old prints stored in a box in a bag in the fridge grew. None of my 9 year old prints that were accidentally stored with seeds in my larder [cool room] grew, even with dense spore spread on agar.
20 Year old well stored spores have been revived.
6 Month old spores that people kept in a shoe box under their bed with a porno magazine have been dead.
Its all how they were treated.
It also really helps if the prints were made correctly, naturally.

Even live mycelium suspended in sterile distilled water has lived 20 years in a refrigerator.
