Today I judged that my agar plates were ready for some transfers. Here are a few things I've learned so far:
* When you see pics or videos of experienced people using sterile technique you think, "OK, simple. Just do X, Y, and Z." But it takes practice. I'm still fumbling around the SAB like I was at age sixteen trying to unhook a bra.
* When the alcohol is inside the box you need it on the outside. When it's outside you need it in there. Genius...what if I had TWO bottles?
* When it comes to inoculating a plate from a MS syringe I truly am an artist. And that artist is Jackson Pollock. Like snowflakes covering a tiny frozen pond, that shit was everywhere.
* When you're carefully lowering your "scalpel" (xacto knife) into the pretty white mycelium it's really disheartening to hear the hiss reminding you you forgot to cool it in a less precious part of the dish. Even more so the second time you do it. And...the third.
* Being baked while you're doing transfers...I definitely made the right decision there.
I'm definitely glad I decided to start using agar. I'm gonna pick up another couple packs of PP5 this weekend. I never thought standing in my underwear and a pair of surgical gloves slicing up gelatin and breathing alcohol fumes could be much fun but I'm really enjoying it.

laughed several times while reading this, this is me to a "t" and youre right it seems simple then you do it and finish and have a list of about a dozen things to do different next time. i would imagine if i had a friend who was a surgeon and i explained the goal he/she would come up with some good protocol / order of operations to go by . good post

Take your time man your gona fuck some shit up at the beginning no matter what, so account for that. I still fuck shit up and I’ve done like 50 plates.. Practise some dry runs if you need to.
You want to learn good habits right off the bat. Watch videos. Read sterile teks. Study like your cramming for your grade 10 math exam. I literally go through the motions for like 30min before I even load the sab.
,..And for the record, I always work in the sab stoned. Keeps me calm and collected.

The SAB felt like an old friend when I started working in one! Maybe it's my time in kitchens with raw food rules and such. I also like processes, so who knows.
Watch out with alcohol in the SAB. If you're properly flaming your scalpel you might make a small bomb.
