Agar transfer prep

Got a plate that is starting show some decent growth from a MS syringe. Is it possible to take 6 transfers off a plate just to get multiple plates going if so could this plate be capable of doing that?

Should I give it couple more days to create more vectors to transfer from or just go ahead?

Nah, you got enough there to tranfer from. Looks like really whispy MS so you should be able to isolate it down to something in 3-4 transfers.

Get to it, the first one is typically gonna be dirty from a syringe so you can start with any growth that has an edge you draw from

Here’s a better pic.

Quick question I could keep transferring out plates from this growth? So say I take 3 plates of this than transfer each those to 6 more, I can continue to create plates so I keep tubs moving through?

Got three transfers knock out. Keep the original around is that a sensible thing to do? More transfers or continued growth?

Since I reuse my petris, I keep them until I need the dish or it has sporulating mold. But it's nice to have it to compare to transfers, it may reveal a contam or it may make pins for more agar experiments.

I like to keep the parent plate around until the child plate is around 50% colonized. If the child plate contams, you can go back to the parent and try another transfer. There's nothing wrong with taking more transfers, but just realize how that can increase your plate demand exponentially. If you have 1 parent plate into 6 children, do that for three more parents, now you have 24 child plates... shit adds up QUICK No more than 3 child plates for me, but you can handle it however you wish

Right I’m trying keep 6 monos going at all times, so I can go through plates really quickly. I like the agar because take the 6 plates into 5qt spawn bags and your ready go to the tub in 2 weeks!!

Yeah I would look into Liquid Inoculation, I am fixing to rewrite my TEK on how it is done using children plates. Basically its about getting some jelly jars and an oyster blender and blending up the children and then spreading it over the masters.

Would Blenderless inoculate work as well I’m trying stay away from jars and just use the 5qt bags so I’m not waiting on different colonization times.

If you take 6 transfers from 1 plate you’re not being selective enough, and if you propagate that to 6 more children all you’re really doing is practicing sterile technique. The whole point is selecting the best genetics from the plate and propagating only the best from there.

It isn't all or nothing though. Take 6 transfers and then be selective with the results. Plates are cheap and don't take much space.
