tbh i think id rather inoculate one master jar with a syringe than with agar

I cant tell if my agar on the bottom is contaminated or if i introduced contams when i opened the jar i think its bullshit when people say that its useful in determining whether your syringe is clean it is indeed but the trade off is a much longer exposure to air and more of it than if i used a syringe for one second all sterilized etc...Maybe if i was selling syringes or making them for personal use which i would because other than using it for cloning i dont see the point

Your sterile technique is bad then. Exposure to air doesn't mean a thing if you doing proper sterile work. You can leave a plate open in front of a flow hood for a whole day without contamination. That's not saying you need a flow either to work well. I'm just saying air exposure isn't the issue. Learn how to work in a SAB properly.
Prints are never sterile, thats why they need to be cleaned up on agar. You put shit in, you get shit out.

I used to think the same way as you, that the more tha jar opened the more chances it has to contam. What I should have been thinking was “the more I fuck around and don’t treat my jars right the more likely they are to contam”
Everyone on this site opens their jars and inoccs them with agar. If they don’t, they will be soon. Inoculating with a syringe is more dangerous, and cancels out whatever advantages you think it’s going to give you.

Are you in the SAB when you opened your jars? Does it get hot in there? How big is your SAB? Oh and Sweat can be a contamination vector if it gets near your jars.

yes was in the sab, i suppose it gets kinda warm i just have lysol wipes evaporating in it, its medium sized. No i have gloves attached to pvc no worry about the sweat lol

detach the gloves and flsme outside the box, i sweat but it isnt a problem and i also dont use gloves at all, learn to keep your hands from going over your work with or without gloves.

I do go over the center of the mouth just so i dont touch the agar to anything, only the first time ive done this, so i guess ill find out if i fucked up

your strong hand is your transfer side. so if I'm inoculating a jar with a wedge my donor plate is on the right my jar on the left, my hands don't go over my open jar.
