Basic Isotopes Questions

I just finished loading my first agar plates. Now the question is Isotopes.... 1. Is that the goal, other than decontamination? 2. Is there a stigmata/controversy against them? 3. If I am able to get Isotopes, is the goal then to keep it going forever through transfers? 4. At a beginner's level, should my goal just be decontamination and then inoculate?

I would go through everything in my signature, all your questions can be answered by researching.
Also, it's isolate, not isotope. Isotopes are variants of a particular element

I believe you mean isolates. And as a beginner just focus on doin at least 2 but probably 3-4 quick 1/2 grain if rice sized transfers from leading edge and try to get quick uniform growth.
Once you think you have clean mycelium make another couple transfers from leading edge yo new plates just in case and then drop a piece to sterilized grains. Once you get a grain har fully colonized spawn it to coir, fruit it, clone the best fruits/best clusters. Then do 1-2 transfers to clean the clone up and drop those to grain, always keep a backup in the fridge in case something goes wrong you dont have to start at square 1 again.

yes your goal should be clean culture rather then isolates, but if you achieve one it'll be all good

OK, so I really goofed on saying Isotopes. But the thing that me started was this article

Now, don't get me wrong - the mushrooms that did come through are big. But more than half of this guys trays did not come through for one reason or another and I would consider him an expert. I think maybe I am missing something, but this is the only article I found that begins to talk about and show Isolates.

It's all the same variety, 10 isolates of PE. Even after isolating you may not have an isolate that has the features you want. You have to fruit them to find out - that's what this guy did.
It's not necessary to isolate down to one sector (one sector isolated, is an isolate). It is what will get you those beautiful tubs of uniform mushrooms though.
Many folk will go agar to grain once they have clean growth, and then clone the best fruits. That's what I'm doing, while finding isolates.

Thanks for the info. I'm a dork and was looking up articles for isotopes instead of isolates. One final question - Should I treat trying to Isolate a Pan Cyan Jamaican vs B+? I just seem to see people isolating cubes compared to pans. Thanks

No experience there, but my sense would tell me that the agar work would be similar as far as isolating it from contams.
