Maple Syrup Liquid Culture Tek 2

A Foreword:
Firstly i would like to say, that i am no cultivation expert and i am not writing this tek to pose like one. I have a lot of respect for the Cultivators (Trusted or not) of this site and am very grateful for their work. I am writing this tek since i personally had trouble finding concrete information about using maple syrup in liquid cultures. What i mostly found were people asking if it would work - no exact measurements or results. I think that what i have here is valuable info for some, since in certain parts of the world fancy stuff like light malt extract is hard to get (and stuff like Karo is sold at fancy "bio" stores for high prices), while maple syrup is readily available.
So i figured that i would type out this technique (and report results) for those who choose maple syrup for their LC.
What you will need:
1. 100% Maple syrup Just make sure that it has no other ingredients than "100% maple syrup". I used grade C maple syrup, but it should not matter.
2. Potatoes I used them to add some diverse nutrients, but you could probably skip them. Some people recommend those red potatoes. I used regular potatoes and it worked just fine.
3. Jar with a lid of your choice After long contemplation, i decided to use a lid with 2 silicone ports. It works fine, but if you want to use this, you have to leave some empty space in the jar, because there is no GE filter (i had a 400ml jar, which i filled halfway up - 200ml LC/ 200ml air). You will also need an airport syringe (syringe without a plunger, stuffed tightly with polyfill) to relieve pressure after pc-ing.
4. Water I used tap water. Some people use distilled or bottled spring water. It may be better, i don't know, but unless you live in Flint (or have really hard water), tap should work.
5. Pressure cooker Some people sterilize LC's in a microwave, but i don't own one. I also read that you can get away with steam sterilizing your lc, but I have no experience with that.
For 1 Liter of the final product you need:
200-300g of peeled potatoes 40ml of Maple syrup.
From my research, it seems that maple syrup is about 1/3 water... Now, people usually make their LC contain 4% sugar (maple syrup in our case). This means, that technically you should add 6% of maple syrup (60ml per liter), but it is important to remember that if you add too much sugar, you will compromise your LC. Since we are also adding a bit of potato (which also contains a bit of sugar) i decided that - "better sparrow in the hand than a pigeon on the roof" - and added only 4% (40ml). If you do not want to add potatoes, then i would advise you to add 6% of maple syrup.
What you will do:
1. Wash and scrub your potatoes really well. 2. Peel your potatoes and slice them into small cubes (roughly 1x1cm or so... it will expose more surface area). 3. Put them into a pot and add your water. (Now, what i did is i added only 300ml out of my 1l, because the smaller amount will filtrate easier. This is not that important.) 4. Bring to a boil and then maintain it for 10 minutes. 5. Strain your potatoes but SAVE THE WATER. (you won't need the potatoes anymore, you can eat them) 6. Filter your potatoe water using coffee filter, cloth, fine strainer or whatever (you dont even need to do this, but the sediment will later obscure your vision a little bit) 7. If you used only a portion of your overall water, you now add the rest of it. 8. Pour it into a second, clean pot and bring it to a simmer. 9. Add your maple syrup and stir until it completely dissolves (it will take a minute or two) 10. Fill your jars and PC for 30 minutes at 15psi (i did 45 minutes, for i only have a 10psi PC)
Done! You have your Maple syrup LC.
After this point, do whatever you want with it (unless you want to knock it up with spores). I will write what i did bellow, so you can better judge the effectiveness of this LC.
What i did with it:
Since i was using a lid with 2 silicone ports, i had to pc my jars with the lid slightly open (not enough for it to fall off) and covered with aluminum foil. At the same time, i sterilized my airport syringe and a syringe filled with water, since i was going to knock up my LC using the biopsy method. After the pressure dropped, i opened my PC and quickly tightened the lids without taking the foil off. After it cooled down, i moved my jars, airport syringe and syringe with water into my still air box. I then wiped my donor agar cultures with ISO and too added them into my still air box. I relieved the pressure inside my jars using the airport syringe (stick it through and wait for it to equalize - this will suck hopefully sterile, fresh air inside). Then i shook the hell out of the jar (to return oxygen into the liquid).
Knocking up went like this: 1- sterilize the needle with flame, then push a few drops out to cool it 2- open plate 3- poke the best-looking part of the culture (now there is a microscopic piece of mycelium in the needle) 4- quickly push the needle through the silicone port 5- eject 1-2ml of water. THE STREAM HAS TO HIT THE LC SURFACE (the water flushes out the mycelium) 6- sterilize the needle and repeat with other jars of lc/plates
I basically used this tek: https://www.shroomery.org/forums/showflat.php/Number/24740168
I "incubated" my LC at about 25-27 Celsius (temperature in my growing space), stirring it 1-2 a day. After 2-3 days a tiny white spec appeared... a glimmer of hope you might say.
Results:
This is how the culture looked 9 days after inoculation:

Strong and fast growth, considering it was knocked up by a biopsy. I decided to not let it grow any further, to prevent it from clumping together. I knocked up some jars and stored the LC in the fridge.
This is how the spawn (0.6 L of rye grain knocked up by 2-3ml of LC) looked like after 8 days of colonization:

For comparison here is a photo of a jar knocked up by a slice of agar, after 10 days of colonization:

Last words: While this may not be the best LC recipe out there, I think that it is appealing none the less to certain people. I personally have used maple syrup lc several times and gotten much better results than with Honey.
I hope that i have demonstrated the effectiveness of this recipe and helped somebody out there who was in the same situation as i once was.
Thank you for reading. Enjoy your days. -Ombisha

hey! maybe its because im in vt, but i just tried maple liquid culture myself with (seemingly)great success so far. i figured maple syrup is already heated therefore doesnt crystallize during pc. i get fresh grade a here , it came out of pc very clear with no visible sediment or floaties. so i did a comparison with honey lc and wasnt even close so far using both wedges and mss. my process was basically pc 1TBS grade a dark amber syrup with <pint of water for 30 minutes at 15psi let cool, inoculate in flow hood let sit at 78f. just did this 2 days ago but already seeing some good colonization in maple, hardly noticeable in honey. good to see the connection to tree sugars getting involved. i am new to the hobby, but old to the sport

Thank you for the comprehensive explanation and illustrations. I have a batch cooking in the pressure cooker at the moment. I've had problems with combining liquid ingredients in recipes before, but I hope this one turns out well. I've saved it as a favorite, printed it out, and bookmarked the thread for future reference
