question on long(er) term storage of myc

i was able to clean up some LC with agar and made some grain master jars but they seem to be drying out and the agar has completely covered the plate. how do you guys store your myc long term to continue growing?

If you are trying to save a culture you can put it to a slant. So basically you would take a small test tube or glass item with a lid. Add agar halfway up let it cool at an angle. Some people use popsicle sticks for wood lovers. The idea is to create an environment the mycelium can colonize that you can put into cold storage for years.

i am semi familiar with a slant, but how is that different from just a normal petri dish?

You can do slants in plastic fine. I use 50 mL centrifuge tubes that are polypropylene plastic and they work great.

Also you do want some gas exchange as well. I give my tube's capo a half turn and then wrap with parafilm. Mushrooms are aerobic organisms so they require fresh air. Your slowing down their metabolism by putting them in a fridge but they're still alive and still "breathing".

The idea I've heard with a slant is you are creating a gradient of micro-climates across the surface. I'm not sure that's really true or not. The biggest factor is refrigeration though. I also add a wooden Popsicle stick and that gives to give long term food source. But hell, I just revived an agar wedge I found in my fridge that was a year old without any issue.

And the difference is that a slant doesn't dry out and curl up after a few months like a thinly poured petri dish does. But you can also pour a thick plate and that way your agar Puck won't dry out as fast and you can refrigerate your cultures for very long time if you insulate the sleeves with a sock or some cloth or whatever (in case your fridge throws one of those freezing moments that usually destroys live cultures)

I make an LC with 1.5 tbsp of white rice flour in 500 Ml of water. Once its ready I put some in a 15 ML PP% or glass vial, with the lid on tight and wrapped in parafilm in the fridge. The stuff is the consistency of thick motor oil. I've kept cultures alive for years at a time like this with no "slow down" (even had a couple times where the thermostat was bumped and everything froze).

A few drops goes a long ways.

My rice flour slurry lasts way longer than my slants or any other agar preparation. Rice flour LC is also my fav for inoculations but I only put 2 tsp per 500 Ml if its going on grain soon.

Even at room temp rice flour LC can last a long long time.

did you just rehydrate it?

im worried my grain master jars are too dry now and i cant use them to spawn more jars :frown:

There absolutely is gas exchange. If not where does all the moisture go when the dish curls up as it dries.

There's not a lot but it definitely does happen.

It was in a microcentrifuge tube and still had plenty of moisture in it so no need.

Yeah but from the context they were clearly saying there wasn't gas exchange. I never mention FAE at any point. Also there's a big difference in the gas echange happening between a threaded container with the threads completely tightened and loose. That's why I do a half turn.

fresh air is made up mainly of the following, all gases:
Nitrogen, Oxygen, Argon, Carbon dioxide, Neon, Helium, Methane, Krypton, Hydrogen, and Xenon

Once again, I'm telling you FAE does not occur in agar cultures, GE occurs always if not hermetically sealed.
That's what I'm on about, showing you that you clearly don't understand the difference between FAE and GE when it comes to mushroom cultivation.
