Pictures of practice run 1st time agar + Bac jars in mono update
    #26489707 - 02/17/20 09:43 AM (3 years, 11 months ago)

Backstory with some detail in qoute or skip right to the TLDR at bottom

Quote:

After all these years of getting away with  MS to WBS and just growing for fruit ive become interested in learning more about working with agar and eventually building a culture library..well once im able to fully understand whats actually on my plates and making optimal transfers etc.

So..since i never done it I wanted to get a feel for making agar, pouring petris, seeing how my technique was innoculating in a stack bottom up etc etc.

Needless to say.. it felt cumbersome at first attempt. Very clumsy. I couldn't feel the petri lid with gloved hands and even poured some agar on a lid of an empty petri by mistake.

I have realized what i need to do to improve.

I managed to get through the process of what i call a "practice run".

The inoculate was a leftover MS vendor syringe containing about less than 3cc.

MEA premix from a vendor was used to create the media and was ran in the pc for 20m at 15psi.

I simply dripped whatever came out of the syringe on the slightest push.

[I have pre packaged plastic inoc loops for next time to use the dip and streak method]

TLDR;

Before you see my wierdo agar below you should know whats in this jar?

Ok , TLDR2

Here is 4 of 9 petri from agar practice run

Help me identify and learn what i have here.. no possible way i have anything usable but i am too much an agar-noob to know.

What Im noticing are these clusters of white with white centers with paler/off color between. Looks strange.. maybe because its 2D.

Please explain what this is indicative of?

Is that rhizomorphic myc growth or deception!!??

Whatever it is.. watching agar is neat..

Finally two last things,

1. The last 2 of the 9 petris didnt get spores because i ran short. I flicked the empty syring over those 2 petris for a "hail mary" play just in a case. They are both looking uncolonized by anything. Just blank agar.

I suppose that can be seen as a good thing. No C word on those two.

2. Several dishes had condensation so as a test i unwrapped the saran from another dish [dish 5] to see what would happen.

I let it sit out until the condensation was about 80% cleared up and then i placed it [ still unwrapped ] in a ziplock bag.

Here is how it looks. Puffy, fuzzy. Def has to be something gross, right?

Both pics are same plate tried to get as close as possible

If you used a syringe and squirted more than I like to. (A single drop)
The fluid will run around the plate and the spores will show mycelium from all different areas. That's normal
Yes your plates look like the growth is a bit odd...maybe it's not mycelium, maybe it is.. let it grow out for a few more days and take more pictures.
The jar looks great. I'll be surprised if that's not mycelium. Looks 100% in a day or 2. Maybe shake and let it recover for a few days then smell it.. the smell is the most important part.

Thanks for the advice and input.

On some plates there was too much fluid and yes it did run across the plate.

Ive ordered a new spore syringe which should be here later this week.

I have 20 new petris and plan to do about 10 with the new syringe to try again. Looking forward to it actually.

Maybe...one of the first 9 will offer something to transfer out to a new plate.. idk. Reading up on what to look for but this tiny 2-D grow space is so foreign to me.

The concept is easy but knowing where to go with whats growing is something i need more info about.

Ok now i have 1 vote odd 1 vote looking good

Are you basing your approval on the info of the thumbnails or the pics in the imgur link at the bottom?

If its looking good can someone chime in and explain why the growth is good and not at all odd?

that almost halo-like ring worries me as i look at this 2-d surface with ignorant eyes. The edges looks so good but feel im being decieved.

Nah that jar is bacterial. It’s cubensis mycelium, but has bacterial infection.

The plates look odd but could just be young growth. Don’t toss the jar, spawn it and see what happens.

Yeah, the jar's not perfect, but good/viable.
I looked at all the plate pics. Besides the obvious look of spore germination, there are also some lovely rhizomorphic edges here and there. My germination plates never start out that clean.

OP is the jar multispore or did it colonize from agar?

For the inquiring minds that might read this in the future via Google search, perhaps in 2034 or for even those with us here today..this problem might become anothers in so tell them why you came to that conclusion so they can learn to spot it what you see.

Weird way to say that but

The grains should be completely colonized. Those few grains pushed up against the wall that (I assume) won’t colonize show bacterial infection.

Those petris are not right.

Why are you using spores in jars if you have agar?

The reason we use agar is to ensure clean growth.

Then you put that clean growth on the seeds.

You may get some shrooms from that jar. You may wanna top fruit to lower risk.

I’d spawn that jar for sure. Millet colonizes weird.

On the plates it does looks really weird, like somebody placed spores in a pattern and then they germinated.

Looks like normal spore growth from what I can tell, you’d need better pics.

Does this look healthy?

If so, transfer out to new plates soon from 5-7 oclock?

It looks like some fuzzy mycelium on agar to me but I suck at picture identification. Often the liquid in the syringes kinda leaves a bit of "film" when it drys and can look like those little circles, the various little separate colonies are forming and will continue to merge together. If you shine a light from the back you should be able to see more of what's going on. I have learned not to use too bright of a light when I used an led lamp to sit them on to admire and it cooked the agar enough to make it turn color but check it out and see.

I had very similar "circles" from just spores with no liquid.

What I would do...

Take your extra dishes and start transferring. Transfer small pieces from right before the outer edge.

I'd take a few samples from each plate and your growth will look much better on "G2".

I wouldn't ditch the plates. As for the jar it's hard to tell. See if you can smell anything through your filter. If its sour you will know quickly...

Im going to make new petris in a couple days and do that.

As for the jar, I shook it one last time to see how it recovers.

During the shake it broke up really easily and smells like mushrooms. No funky smells noticed but will follow up.

Getting this really fluffy 3D edge

All plates are still without discoloration and are whiter than when first posted.
