Bacillus Vs. Mycelium on agar question
    #26489109 - 02/16/20 09:33 PM (3 years, 11 months ago)

Has anyone ever tried refrigerating an agar plate after inoculating it with contaminated spores?  At room temp it seems that infection usually dominates before the spores have a chance to germinate. My question is, would a cooler temp slow down bacterial growth just enough for mycelium to gain a foothold on the plate? or would the fridge slow both bacterial and mycelium growth at en equal rate?

Full on refrigerating (3-6C/37-42F) will slow down both way too much. That is how cultures are stored on slants (extra thick agar layer, stored at an angle for years that basically stalls the growth to a halt).

It is true that the room temperature on the warmer side (25C/77F) can help some contamination spread faster than mycelium but the bottom line is pretty clear - if you have a contaminated plate you want to transfer from it as soon as possible to a clean one and grow that out (quite often multiple transfers). So I would not worry about temperatures but rather try to transfer as soon as possible to make it clean. If there is any kind of contamination on a plate (even not touching mycelium growth) - you almost never want to inoculate grains with it, only transfer once more.
