How to recognize an isolate and stop doing transfer?

I'm learning with agar dishes.

I used some spore and put some right in the middle of a dish. A few days later, myc started to grow evenly (circle shaped).

I took some pieces from the outside and put them in other Petri dishes. I now have new Petri dishes where myc grow in a circle shaped way.

I understand the concept of transferring to isolate genetics. But when do I know when I got an isolate? It seems to grow evenly. Do we just "assume" that it's an isolate after 1 or 2 transfers?

Basically: when do I know my colonized agar is ready to go into a jar?

The growth is good to go in a jar when the growth is a nice circle and preferably rhizomorphic. That way you can be more confident of no contaminates. Tomentose growth could very well be cube much, and probably is, but harder to tell for sure. Uneven growth (non-circular) could also be not contaminated and just different genetics, but there is a chance it could be a contaminate. With Rhizomorphic, circular growth, you can be very confident that it is not contaminated. That being said, I've used uneven growth plates and went on to a wonderful harvest.

As far as isolates, that takes much more time and experience to differentiate strains on agar. And even when you do get an isolate, you must determine if it is one that produces. You may have to grow 10 isolates to find a good strain. That's a lot of work. Being that you are just starting out, don't go for an isolate, just go for a clean growth.
