Frustrated with agar - potential solution!
    #26487940 - 02/16/20 07:13 AM (3 years, 11 months ago)

This hobby requires massive patience, you all already know. I had such amazing luck with PF tek on my first ever go (no contam in 6 jars) that now I'm struggling to germinate spores in agar its a blag. I'm not even going to bother posting more pics because there's nothing interesting on any of my plates. They all look the same - snotty.

I've ordered some plastic inoculation loops and I'm hoping this will improve my luck strategy. To be honest, I had the same problem growing E.coli in the lab once, the colonies were all on top of each other so it was impossible to isolate any. I fixed it by learning proper streaking technique. If I streak the spores rather than a drop from the syringe hopefully it will create some distance between the mycelium and any bacteria, and give me a chance to rescue the culture.

Now that I think about it, this is exactly what I should have been doing to clean up the syringe but it wasn't emphasised anywhere. I just saw recently Bod encouraging use of a loop for spore syringes and it all clicked into place.

Also I think I will try some different recipes/colours of agar. The grey activated charcoal agar looks cool, even if it doesn't confer any advantage to the mycelium.

On a positive note, I had almost given up hope on my latest PF tek where I used an old dry GT spore print, but theres some spots of myc in one (just one!) jar. Even if I can't clean up the dirty spore syringe, could I transfer some mycelium from a brf cake to agar and then take things from there? Should I wait for the whole cake to colonise and then take a chunk after its been birthed? Very exciting if it is cube myc, less exciting if I end up putting mould to grain

Love you guys btw xx
