Liquid Agar to petri
    #26486168 - 02/14/20 09:36 PM (4 years, 2 days ago)

My first time... Within a week I am  getting 10cc syringe  of “Golden Teacher.” My understanding is 1cc is enough to create mycelium in Petri dish. If my math is correct that should give me 10 plates? Or Should I rid of Petri dish approach and just inoculate  the  10 1/2pint jar of substrate instead ( and take a sample of the mushroom afterward? Is there a best approach to this that will give me more spores done the road and better yields?

Also there was this video on making agar in a sterile tub with two hand holes and a air mattress air pump to filter the air in the tub. This guy was an English man, seems like an older video. Anyway I can’t seem to find it. I’ve seen videos on this but this seemed like a good approach. Thanks in advance.

Ps I would like to create a thread in which I document the process and get feedback from experienced growers. Best place to post to get immediate and good feedback

One drop is enough to do a plate. I would use as little liquid from a syringe as possible.

A still air box is what you want. Anything with moving air short of a true HEPA flow hood is bad news.

You only need one or two drops of spore solution for each Petri dish. You can either put one or two drops into the center or the agar plate and let it colonize or use an inoculation loop to spread the spire solution across the agar.

If you’re looking to get the most from that one syringe you can’t beat agar + LC (liquid culture). I’ll link BOD’s agar guide below.

My thoughts... is this the still air box approach⬇️⬇️⬇️⬇️?

Two holes for hands and for the back cut out a hole for a very small fan and put hepa filter in front of fan. This would allow for no dust to fly into my agar right.🤔

Don’t over complicate it. Burn two holes for your arms, spray inside, let it settle, and do your work. You’re going to be more likely to introduce contaminants going through all that extra work than you would be keeping it simple. If you want something more than a SAB save the money and get a proper flow hood.

The point of a still air box is to STOP air movement. That totally defeats the purpose. Unless you can have sterile laminar flow, you want a still air box. To build a proper laminar flow hood cost at the very least a few hundred. If it was that easy to get sterile air everyone here would do that instead of spending hundreds of dollars on flow hoods.

No. That video those guys made is wrong on all accounts.

Also, one drop per plate is plenty.

OP - Be sure to read the stickies in getting started. The ones related to cultivation are in my signature.
