Agar transfer or remove contam?

Hi all, I tried searching but couldn't find an answer to this.

I think I may know the answer already, but still it doesn't hurt to ask, right?

I have an agar plate from a spore print ( my SP technique was not great at the time). The spores are germinating, but I see a spot of what I think is mould on one side of the dish.

Should I:

A) transfer healthy material to a new dish (means I will need to pour some new agar)
or
B) remove the contaminated section

I'm guessing the answer is A, because disturbing the contamination could spread it.

Also, how quickly do I need to act? The mould spot is about 3mm across and looks furry. Do I need to be very careful in handling the dish so as to not dislodge and spread mould spores?

What say ye?

I mean, if you're worried you could toss out the plate and start over. But that would be a waste of mycelium.

I don't think anyone here would suggest removing the contaminated section. There are likely other spores, and, like you said, moving the mold will likely contaminate the rest of the plate anyway. Transferring agar from the cleanest part of the agar furthest away from the mold seems to be the best option.

And ideally you'd act as quickly as possible, given how fast mold grows.

Yes, I'm quite keen to preserve this one as it's a strain from which I only took one spore print!

I also want to at least attempt to clean the culture in this way rather than just giving up on it.

So I poured some new agar plates and transferred the good stuff into three. Two days later: apparently we have success! The growth has continued free from contamination and so I have preserved the strain I did not want to lose! (Treasure Coast)

I will store it in a slant for now until I am ready to cultivate.

Very pleased with the result of this. Now I understand the power of agar work. :smile:

Congrats, dude! Just keep watch for any contams that might have hitched a ride but haven't shown themselves. It's taken me 3+ transfers to get something actually clean, when I thought it was clean the first time.

And yeah, agar is so nice. You can take the dirtiest source, and get something sterile out of it after a few transfers.

What if the spot of Fuzz was just spores that fell during plate inoculation? Doesn't matter cause you want to isolate genetics down, but what if.......

I had multiple spots of growth as I dropped spores from the print widely across the plate. It was pretty clear that the growth was different in this area from the others. The growth started out more furry with a different texture and wasn’t spreading in the same way. I think the best way to tell is that it just looks different. Now after a few days I can see two spots of contamination and both have turned clearly grey-green with a yellowish fringe.
