Agar inoculation question.

When inoculating agar with an spore syringe, do you pierce the agar with the needle or simply allow the drops to land on top of it? Also, which method is best for inoculating agar, spore syringe or spore print?

Most will prefer prints as it is at least one less contam vector. I think it's up to you how you want to get the spores on there. Just remember that less is more, especially with syringe.

Just put a drop or two on the agar. This can be kind of hard in reality so don't stress too much if you squirt a bunch out.
a print is usually better because you can swab directly from it. That means less contamination vectors.

And you just drop what you collect from the spore print with the scalple directly onto the agar? None of the zig zag Z figure with an inoculation loop?

A scalpel isn't ideal but it'll work. I would use sterilized q-tips wraped in foil. the pattern isn't super important, but the zig-zag covers more area and gets more off the swab.

Zig zag is recommended to spread them out. But just getting the on there will work. Just nice to avoid 1000's of spores germinating in one spot.

Could always just try a couple different ways, ive done straight scraping a print a very small bit with a scalpel then just pokin the tip(just the tip i promise)xD into the agar in a couple places. But lately i just fashioned my own inoculation loop out of a christmas ornament hanger into my xacto handle and its been workin great for me to do the zigs and zags

Cheap n easy and you may have a box of em in your house haha Same concept just flame er red hot poke in agar to cool then scrape a tiny tiny bit of the print then do your pattern
