How should I seal off my test tubes for coffee experiment

Im preparing to do my coffee experiment tesing the growth allong tests tubes after doing a G2G into each tube here is the post showing the basic procedure http://www.shroomery.org/forums/showthre...amp;o=&vc=1 Now im debating over if the polyfill with foil is the best meathod. I was playing with grain in the tubes to see what it would be like to try and get consistant filling and it wasnt working that well. I was thinking how much easier it would be to measure the clean grain going into the tubes with a leveled measuring cup, then steralize. Now when i G2G i planed to transfer and leaving a 1" space from the top of the grain to the rim so theres room for polyfill. Instead id fill the spawn till its overflowing then with a sterile knife level the top and re-install the coffee filter. This should give me a more consistant spawn amount making it a bit more of a controlled test. Now my question is will a coffee filter with foil just for steralization be enough to stop all contams, or should i stick with the pollyfill?

coffee filters won't do .
but, you could use tyvek, or a band-aid .

Thanks! I can get a new tyvek suit from my dad since he does Hazmat removal and cut that up. Then just rubberband or twist tie piece on top.

Since the tubes are pretty small I'd use a filter, if any, that will help to reatain the moisture on the inside of the tube.

but that wont let it breathe. Having a 1.25" spawn layer on top im not worried too much about contams. Ill be conducting this experiment sometime next week. My jar is 80% colonized. Im also removing the caffine pills and just doing one test group of unsoaked CC since i broke one tube and another i filled with BB's toped off with hot glue that wil be used to measure the grain to fill thetubes.

Parafilm will breath. It is often used on cultures.

Ok.. well i already have the tyvec. If i have problems with the tyvec ill buy some parafilm.

If the Tyvek is without holes, it should work.
I've seen Tyvek with minute perforations in it to aid in respiration. This stylr Tyvek would probably dry things out too fast.

I ran out of quart jars and needed to start a few more strains so im going to the test tubes. I have the pc cooking as i speak. I preped 9 tubes with plain wbs. I left about 1" at the top and stuffed a polyfill wad and wraped with foil for 8 of them and the 9th one i used the tyvek with a rubberband and foil. Im going to be using them as individual g2g tubes and just plop a whole tube into a preped q jar, once i get more free jars.
