What kind are you using? I would suggest going to your local hardaware store and buying a roll of blue painters masking tape. It works well,as it is porous, and it will hold up to the pc at 15psi.
Good Luck MYKEY

You might try real duct tape (the metal kind). I have done this anytime I have needed to tape something that goes in the PC. It has worked extremely well. This stuff is very strong and comes in very handy. Edit: I never actually tried to poke a needle through this stuff, but I bet it wouldn't be that hard. It will seal very well though, it does not come off. Pressure cooking it seems to make it stick like glue...

I KNOW that you can not use the sliver duct tape. I have attempted to used it and it melts... I dont know if the metal duct tape is different (I am assuming so). Normal duct tape melts at 93 degrees C. The inside of the PC at 15 psi is 121 degress C.

Masking tape survives for me.
Gr0wer, its for the holes on PF cake jars. Beginers dont usually use Tyvek or Polyfill, they usually do dry verm layer and tape

I used tape to seal the top of a small vial I was sterilizing that contained rye extract (for agar). The plastic would melt, so I used tape. Tape is also commonly used to seal the holes as in with the PF tek.
The metal tape works very well, although it can be expensive.

Micropore tape withstands PCing and allows the culture to breathe. You should be able to obtain it at most pharmacies.

Go to Lowes/Home Depot or somewhere like that and get aluminum tape. It's called HVAC tape. It'll hold up. It's expensive as hell though. Here, in Texas it was $13 a roll!

Same here. I've used several brands of black electrical tape, and they've all worked well. I used them to cover holes on PF jars.

3M micropore tape works very good , as far as masking tapes go , only tape for autoclavs will hold up , but you must make sure to keep tape above water line , this tape is costly and changes color to let you know if you have achieved proper sterilization temps . Good Luck ps duct tape will leave glue residue inside your pc , possibly even goo up your pressure gauge .

Sterile cotton(stuffed in opening ) with small chunk of foil over top works well for what you are doing . Cotton makes good contam barrier , foil prevents cotton and grain from getting overly soaked .

polyfill works better then cotton since it has no nutrients for conams to grow on.
