Temps are fine I would keep it around 75F if you go much higher you will be giving contams an environment they can thrive in. Much lower and growth will slow. Id say keep it 70-75°F

What now? Best pics I could get through the pastyplate:


With a good visual through the container I don't see any fanning or sectors, just a white fuzzy donut. It also looks clean as can be. Should I transfer a couple chunks to some more plates? Is this plate good to go to grain after a little more growth?

Easy as fuck Brown Rice Flour Agar
6g BRF 9g agar 400ml tap water
boil till dissolved pour into media bottle PC 15psi 35 minutes
pour or no pour

Post in the agar envy thread. I don't like how high it stands off the surface but it may be fine. I posted a plate that looked almost like that a while back and was told it was fine. Not sure what causes it but when I get growth like that I keep transferring. Whispy growth is fine but I don't like fluffy growth. It may be ok too but I just don't like when I see it post on agar envy and you should get help faster than here in this thread.

How long did it take to get that growth? How are you storing it and what temperature?

Hey gizmo, just an update on my agar. It's been abut 5 or 6 days in, still no sign of anything, my drop of inoculate from the spore syringe is still there. Do you think it will fair me better to sterilize a blade, and then pick a grain out from a jar that I have colonized and put that on agar? I should definitely get some growth from that pretty good correct? Nothing has happened yet from me inoculating my agar, and I'm eager to see what's been fucking my B+ up and to transfer it out of there. Does that seem like a better way to do it, cutting a piece of colonized grain out from a jar, and putting it on another plate of agar?

Yes you can definitely drop colonized grain to a petri. I have some long tweezers I use for this I think they came from Dollar tree in a 3 or 4 pack where the tools are at. Anyways I just flame the tip of the tweezers then either let them cool off in the sab or you can cool them in the agar. Then use them to grab a single colonized grain and drop that to a petri. Or in your case a no pour.

When doing agar is the a best optimal way of inoculating? For example, should you just use a drop of MS and wait? Or is the streak method the best? And what is the best way if using prints?

I like to make my agar more watery when using prints, they will germinate faster.
I use 9g of agar agar instead of 10g for 500ml.
Scratch it with the inoculation loop and use your preferred streak method.

Streak is probably better you get more colonies to chose from instead of all the genetics being in one place. I don't do that though because I don't test multiple colonies I just use the fasted looking growth and put it to grain as soon as it looks clean and uniform. I then spawn and pick the best looking genetics from the fruits in the tub. I clone them put them to grain and test each clone to find my keeper out of the bunch. To use a print you can either use a sterile swab to swab up some spores and streak them to agar or use whatever utensil to scrape some spores onto agar. My chosen method is to scrape some spores onto agar with a homade ass loop. Somes I'll just use my exacto though. You can also make a syringe but imo syringes suck for agar.

So after a week of inoculating my agar with a drop of spore solution I can see a very faint light halo of white fuzz around my drop. It's almost impossible to see with the naked eye, I thought it was still doing nothing, but I happened to notice it when I slightly angled it and held it up next to a window. It's nothing I can take pictures of yet, and my drop is still clearly visible. Is this how the growth starts and will get stronger? Or is this already a sign of something indicative of my culture? It doesn't look anywhere near the starting pictures I've seen where there's an obvious growth of fluffy whiteness and mycelium.

I'm guessing it's just new growth and with the extra moisture from the syringe it's going to look weak and clear at first. I'd go ahead and transfer it to another plate to see what you get. Mine always start out looking like that too but after the first transfer or after letting it grow out awhile then it looks 👌.

The fuzz is literally so small and minuscule I don't think I could do a transfer of it right now. I'm going to wait another couple of days and hope it gets stronger and spreads so I can cut a piece to transfer. With how small it is I think it would be almost impossible to cut a piece that small to obtain what little there is. Unless there are some tips and tricks that I don't know?

You say halo, that could be anything from myc (cube or mold) to bacteria
Based on you saying it's around the place where you put your spore solution (rather than INSIDE the drop), it sounds like bacteria.
But cant know without seeing it. Use a backlight and it should be easy for us to see

I believe a backlit plate pic can be helpful only if presented with a well lit surface plate pic.

A lot of times there’s growth even when you can’t see it. Like when taking transfers from a normal plate you can cut a wedge from the area right outside the growth that looks bare and it’ll still have myc in it.
If what you’re seeing is cube myc there’s a lot more of it than is visible.
