A couple things questions so I know for sure when I do make it this weekend:
1) Do I need GE holes at all? Or just straight unmodified top? 2) When PCing, do I need to leave the lids really loose? Or do I tighten them down? 3) Do I just store these in a cabinet somewhere when done? Or do they need some special storage?

What is the best ratio you have found to be the best for inoculating agar? How do you make yours?

I’ve never done syringe to agar. Usually I make it softer for spores but you won’t need to since yours are already hydrated.
I’d prob go 9-9 to 500ml. Really anything should work.

1. Depends on the container if you get something with a screw top you can just leave the lid loose. If you use something like glad mini rounds you will want a hole so it doesn't collapse in the pressure cooker. 2. Also depends on container I like to leave mine loose. 3. No special storage needed in a cabinet is fine.

Gotcha. If I decide to go with the mini mason jars should I turn the top of the lid upside down to prevent a seal from occurring?

Yes. If you are going with the metal lids flip them and tighten very slightly but dont over tighten. If you go with plastic lids screw them down until they stop then back the lid off a quarter turn or so.

Do I need to put a hole in those lids like we do for grain when in the PC? The pressure won’t shatter or break the jars?

As long as the lids arent tightened all the way down it'll be fine. Metal lids flipped wont seal and creat a vacuum and plastic ones likewise as long as they aren't tight.

I’m going to piggy back on this thread with my agar questions as a noob. I’ve read the holy grail petris and will likely hop on board in the future when I can absorb the shipping timeline. The route I’m going right now is PastyWhyte’s no pour > BOD’s oats > tic tack toe tiger drop. Some questions that I appreciate any info on below:

I’ve been doing brf cakes for awhile and my timelines have been way off in the wrong direction from what I’ve read on this board. I’ll be moving places in 3ish months and would like some opinions if it’s possible to get a flush within that timeline. My house is above 70 degrees.

Three months is definitely doable.
If the main goal is beating that deadline and you have a syringe I would do a quart or two of syringe straight to grain along with the agar. Starting from spores you’re looking at around 3 weeks minimum to have a good plate grown out for grain and could easily have a shoebox spawned in that time from syringe to grain.
Once the plate is growing you usually look for uniform healthy growth and then take the tip of whatever part is farthest along. Don’t worry about trimming off the agar just put the whole piece on a new plate.
I’ve never used oats but they’re kind of controversial around here, a lot of people have bacteria problems with them. Wbs is probably the easiest, although that’s all I’ve used myself so I can’t say for sure.

I love the tiger drop thing with the pasty plates. I'm using it for grain transfers and it works awesomely. Limits a lot the probabilities of getting a contamination.

So are you saying to double down, do plates with MS for insurance, and do straight to grain jar for speed? Either way, do you have an opinion on volume dropped into both mediums?
Oats are super easy to come by where I live, I wasn’t aware of the controversy. I’ll read up on on the wsb tek.

Syringe to grain 1cc is the norm.
You could get really lucky, my first grow I started a bunch of syringe to grain jars and one was done colonizing in 9 or 10 days. 3 weeks is more likely though.
Tiger drop makes me want to make pasty’s. I was trying growing plates out about that size then cutting a circle around the myc and scoring the tic tac toe pattern only part way through the agar with the goal of popping the whole thing in as quick as a pasty then having the scoring break apart while shaking. Turned out to be too hard though.

So another thing I see is that once you’re agar plates done, do you turn them upside down? What is the purpose of this?
Whenever I inoculate the agar should I also then turn it upside down?
So there are no special precautions to take when storing agar plates before inoculating? You can just put them anywhere? Does the same go for whenever I do the inoculate them? Or special considerations need to be taken? I was just planning to put them in a cabinet once I take them out of the PC and also whenever I inoculate them.

I store my plates upside down after advice, this is because of the condensation. While it hasn't helped it clear up much, it does keep the potential contamination away from the agar surface. I'd imagine this plays a part in general storage as well.

If they are wrapped then you can store them anywhere keeping temperature in mind. If you are making plates you don't plan to use for quite a while you can store them in the fridge, however, a lot of people don't feel this is necessary.
