I always use the wedge in the middle, never even thought about it.

Usually I take such small transfers it’s barely noticeable anyway.

Do I need to flame sterilize the blade if inoculating from the same agar plate? I'm splitting four quarters into 2 jars, so a half per jar, do I need to flame sterilize the blade in between each piece of agar no matter if it's for the same jar? Or only flame sterilize when changing jars? Or between each and every single piece? Or I don't need to flame sterilize when dropping agar in and inoculating?

You dont need to sterilize the scalpel between jars when a2g from the same culture/plate if your clean technique is good

If you are worried about picking up contams on your scalpel during the drop, then you can flame it between transfers to be extra safe.

Me personally,  when I a2g I usually use the whole plate (or 90 % of the plate, avoiding edges , if its fully colonized)  for one jar to speed things up. and I cut it into a dozen or so tiny pieces for more inoculation points within the jar

When I do a2g, I am usually preparing to do g2g, so I only do 1 jar and try to give it every advantage to be faster, so I can speed up the time it will take to get to g2g

I'm going to be doing 2 jars, so do you think 2 quarter pieces per jar is sufficient enough?

Well I knocked up my couple jars of the AA+!!! Whenever I was shaking to get the pieces in good placement in the jar, some mycelium rubbed off the agar pieces while shaking and jiggling around, and looked a lot more bare than when it was all rhizo when I put it in... Is that going to affect anything in any way? I’m worried because the mycelium rubbed off whenever I was shaking and jiggling it around.

That's what you want to happen mycellium will start on the grains that it rubbed off on and will also recover and spread from the wedges.

I am a little worried because I have a little condensation in my jars, either from the PC or cooling. But I did everything in SAB with proper procedure. I'm worried the condensation is going to get the best of me if harboring bacteria or contaminants. Usually my jars never have condensation, but I took them out the PC a lot sooner than I normally would have. Should I be this worried and concerned?

P.S>. I like the new avatar. Very "Fight Club" esque.

Condensation won’t cause contams it can only make anything already in there worse.

But it’s normal you’ll always get condensation especially in the morning when it’s colder the heat from the myc will cause it.

Shake or not to shake yet?

I know the one on the left is definitely ready.. but I'm not sure if the right needs some more time because it doesn't look as far along colonized. However, you can't see the second piece of agar in that one, but it's in there. It must be deep in the middle or right behind it, so who knows how much more colonized it is inside...? I wanted to use both of these in one shoebox so I hope they will be ready together. Anyway, should I shake the one on the right too or needs more time?

Don't worry about the agar just shake to break up the myc that's there and distribute it this is mainly what is going to colonize your jars now where the agar wedges end up in the jar is insignificant at this point.
You really tend to overthink things huh? I'm the same way more than a pinch of OCD to my formula. I always thought about it like this, its better to overthink things than under think. Rather this holds true or not I couldn't tell you but makes me feel a little better about myself.

I'm trying to whip up a new batch of agar and my MEA this time is somewhat clumpy and sticky. Is it still possible to make agar with this? Or will it greatly affect it and cause any problems and I should throw away and buy a new one? I'm having trouble even weighing it out and getting it in my beaker even with a funnel, and getting stuck everywhere. It was never like this when I first got it, it just poured easily like powder. If I'm going through all this trouble I just want to know is it going to affect my agar? Or do I need to wait and get a new bag?
