Can I use a wedge from an agar plate, seal then re-use at a later date?

I’ve recently started growing Oysters at home and have done my first agar to grain transfer and all seems pretty good. Only have one of four jars that have contaminated. I have a lot of mycelium in 4 plates that I’ve closed up again just to see what would happen and there doesn’t seem to be any contam, they’ve just kept growing. What would be the repercussions of using them again either for an agar to agar transfer or even another agar to grain transfer? Could there be hidden contams or would the mycelium be less viable?, etc..

Thinking about it I guess I should just forget about them and just go with grain to grain transfers so long as my jars are not contaminated right?

As long as you didnt introduce contaminates, sure you can keep plates. I assume you wrap them with film? Just throw them in a ziplock and refrigerate for future use.

Just keeps them clean in the fridge. Lots of stuff growing in some fridges. Keeps drying to a minimum also.Not really necessary, just personal preference.

That has always been my thought exactly...I keep mine in plasticware as well.

As long as they remain clean, they should be fine. I do recommend doing some transfers to new plates though. That way you can preserve a known clean culture for future grows.

Thanks for the info 👍🏻 I just actually have agar jars rarther than plates which have a fair amount of air volume so I didn’t need a “breathing hole/tape” and they turned out fine, no contams should be able to infiltrate the jars as they are sealed. Slight issue with condensation but I just put them on their side if they start pooling water.

Stored plates or slants do not need a "breathing hole". Next to no air exchange, especially when refrigerated. They basically go dormant.

parafilm SLOWS evaporation, it doesn't eliminate it.  Plates dry within a few months to a year.  And it can get tiny breaks in the plastic from stress of stretching and/or being knocked around.

Ziplock gives 1 more layer of protection...

i dont personally "double" bag stuff after its wrapped.  that being said I have seen sometimes the parafilm dry out a bit and rupture but even then if I am not rustling the dishes around a lot I dont have too many problems. Maybe for long term storage in the fridge it might make sense tho.  Ill be keeping that in mind.

I don't double bag either, but my fridge stays pretty clean and everything gets an ISO wipe down before it goes in the SAB.  I have problems from time to time but I dont suspect its from that.
But as others have said as long as the plates remain clean after taking out a wedge there is not reason not to continue taking samples I don't think.
