Agar Strain Question

Hopefully this post is in the right section. I've been reading posts on this site for years and did a PF tek grow a few years ago. I just started a new PF tek grow to hopefully produce some nice shrooms to enjoy with my girlfriend (she's never done them so I can't wait!). I've only tried them a few times so I'm looking forward to it as well. Anyway, on to my questions:

I've read a lot about using agar to isolate a strain and to ensure a clean culture. I just used an MS syringe to inoculate my PF tek jars (B+). Hopefully I didn't screw up too bad and I get some decent flushes. I'm planning on taking the biggest shroom I see from a cake with a nice flush. I apologize if I'm screwing up the terms/names. From there I'm going to cut off some tissue from the stem and place it on an agar dish. Then I'll do the whole transferring deal where I take a portion of the rhiz (I think that's what it's called) and transfer it to another place until I hopefully have a plate filled entirely with rhiz growth.

From there, do you keep this particular strain going forever? As in, use this final agar plate to create LC or more PF tek jars/bulk? Or would you just use this agar plate to grow more shrooms and if you find a bigger specimen then you take a portion of it and start a new strain isolation? If I keep using the same strain, will it eventually become so inbred/isolated that it will become a shroom version of Deliverance with crazy ass deformities?

Hopefully someone can help me out because I'm driving myself crazy trying to figure this out.

You can do that yes take your biggest baddest fruit and take tissue to agar then you have a cloned fruit! (transferring upto a few times to ensure its clean)

The mushroom that you clone could be either rhizmorphic or tomentose mycelium (ropey/fluffy) and you don't know until its growing on the plate... Doesn't matter though as its proven to be a good fruiter and the genetics of your clone do well in your environment (different strains like different conditions, some will be more resilient to temperature changes ect)

Once you have the clean clone culture on a plate, take say 5 transfers to 5 clean plates... Keeping the original master plate in the fridge and letting the 5 new transfers grow out then use those to inoculate anything you like (grains, brf, LC).

Once those 5 plates are used up goback to the master plate in the fridge and take 5 more clean transfers to 5 more plates, once the master plate(in the fridge) is running low on mycelium take a new transfer (or two) and create a couple more master plates... Repeating the cycle.  This will keep the genetics young and vigorous. If you were to take a baby from a womb and place it into another womb and keep doing that never letting it 'birth' youll eventually create a hella ugly monster noboby could love.

As you said you could use multispore to find an even more badass fruit then the cycle begins again :smile:  look into slants for long term storage of a culture but if your planning to be growing it alot just keep em on plates.

Oh I see. So isolating a strain doesn't eventually make it weak as long as the mycelium isn't too old? I know I'm thinking too far ahead but I was just wondering if there's a potential future downfall to restricting all your grows to one strain that you've isolated. Although I guess spores from a shroom, even if they were grown from an isolated strain, would still be considered a MS. If my agar experiment works then I may never buy another MS syringe again. This mycology stuff is fascinating! Just when I think I understand it something makes me rethink everything. One step forward two steps back.

Well we're not isolating a single strain we're cloning a set of strains that makeup our chosen mushroom proven to be big bad and fast.
Keeping the master plate in the fridge stalls growth keeping it young and healthy the mycelium isn't working out and getting old, kinda like hibernating I guess.

I think eventually the mycelium would weaken as over time it's still growing although very slowly, but just doing a multispore run or some spores to agar work once a year or so you could refresh the cycle and start a new clone or isolate grow.

Once you have fruits take some prints and make syringes. Syringes to BRF cakes and take spores to agar as a side project if you want.

Spores do carry some traits from the parent fruit like albino a+ produces 99.9% more albino fruits is an example.. But size and potency of each fruit will vary :smile:
