My SAB technique, advice or input?
    #26462471 - 01/31/20 04:54 PM (3 years, 11 months ago)

I will be inoculating my jars later tonight in my SAB I made. I wanted to share my technique of what I'm actually going to do, and you guys tell me if anything is wrong, good, etc. I'm mentally preparing myself and thinking ahead about the work I'm going to do and how I'm going to do it.

I took my jars out of the PC last night so they could completely cool, and I also took the foil off to prevent any condensation build up on the polyfil  from the PC cycle.I've been at work since this morning, so they've been sitting on my coffee table with no foil and just polyfil lids for about 24hours.

I'm going to wipe down the whole surface with 70% isopropyl alcohol. Then I'm going to wipe down all my jars and top of the lids with isopropyl alcohol also being careful not to touch the polyfil. Then I'm going to completely wipe my SAB down the same way with isopropyl alcohol and then put my jars in the SAB. I'm going to wait an hour to let the SAB settle and then inoculate each jar with my syringe. In between each jar I'm going to flame sterilize my needle outside of the SAB.

How does that sound? Sound good?

I have a couple of quick pointers for you for your next run and this current one in the SAB:

1) Leave your jars in the PC next time if you're doing grain. By the sounds of polyfil in your jar, it seems to me like you're doing grain and not BRF. If you leave them in the PC, they will automatically be less susceptible to contaminants. I sometimes will pull mine out early as you did, but if I do, it's because I know I will be working with the jars within an hour of time and even then it never feels right.

2) You don't have to ISO every jar. In fact, this may actually do more harm than good. I mean, you can try it and see for yourself. If anything, I always ensure the lids are more dry than anything so that when I remove the lid, there isn't water somehow leaking into the jar, itself.

3) Don't wipe down your SAB with ISO unless your looking to hurt yourself. Just get a bottle with warm soapy water and spray it. Wait 10-15min and get after it. No need to wait an hour, that's definitely overkill. Also make sure your SAB is in a good spot without a lot of airflow. The more still, obviously the better.

Hope that helps. And yes, flame sterilize between inoculations.

1) Well I was being told in another thread that to take the jars out immediately after they're done and to take off the foil because the condensation will get get on the polyfil and can cause bacteria/contamination issues.

2) I'm not going to be removing any of my lids because I am going to inject through the polyfil, so nothing is going to be leaking into the jar. I thought wiping your jars down is just basic best practice to get it as clean as possible and kill whatever might be there, the same with the lids since that is the inoculation point. Iso dries fairly quickly so the lids will be dry after a wait. Or do you mean that iso is just does more harm overall?

3) How will I hurt myself by using iso? I'm going to let it dry completely, and I'm not going to be lighting a flame inside the SAB. After I inoculate each jar I'm going to flame sterilize outside the SAB and then go back in.

Injection through polyfil is a possible but sucks. Your needle has to be cool first from flaming or it will melt to the poly. If your poly is stuffed tight enough then jabbing the Needle in without pushing the poly into the jar is hard

But afterwards we told you its fine to leave foil on and to leave them in the pressure cooker overnight. One of the people that told you this was a trusted cultivator. You should have took the TCs advice over a random persons.
Also save yourself the trouble and just open the lids to inoculate. This should be done inside of a SAB.
