Slow agar colonization
    #26461244 - 01/30/20 11:29 PM (3 years, 11 months ago)

Hey all,

I'm onto my second grow now, having learned everything that I know from this forum.

I decided to teach myself agar work first and I've been doing pretty well so far.

The only problem that I'm facing with it is that my plates seem to take way longer than they should to fully colonize. I laid the spores on my current batch on the 13th and they're just about finish colonizing the plates now. That's 18 days.

This is my set up variables:
-pasty plates made from local containers (no mini rounds here).
-LME agar made of 500ml water, 10gm agar, 10gm malt extract.
-The weather stupidly humid (at least 90%), with temps up to mid 30C.
-Plates have been sitting in my SAB in a room that gets a lot of afternoon sunlight.

I've only had 2 visible contams from the 30 odd plates that I've inoculated but I suppose there might be something I can't see slowing them down as well.

Does anyone have any tips on what might be slowing it down?
Thanks in advance.

So I didn't finish reading but... dont wait for plates to fully colonize transfer once you have about a quarter sized spot of growth. Then keep going until you have nice uniform and clean growth.
Then take it to grain.

Edit:
Finished reading. Your nutes are a little high. I'd back it off to atleast 8 grams malt extract. I like to use 8 grams agar 8 grams malt to 500ml water. Lower nutes will cause the mycelium to move a little faster.

Have you taken any tranfers yet? I think once it reaches the edges of the plate it loses a bit of vigor.. as gizmo1 mentioned, usually best to make some tranfers from spore plates as soon as you can cut a few small pieces out..

Perfect. You've improved my technique and saved me some time as well.
Thank you so much for the quick reply.

No transfers yet. I've literally just finished making the plates for them. It should be okay to take transfers from these slightly old plates, right? Or would it be better to make some fresh ones?

It should be fine, as long as it's clean

I agree.
Id change the agar recipe then do a transfer if it still moves to slow then start fresh from spores. Or if you already have your plates made up and dont want to waste them just keep going with those until you're next batch. Should be fine.

Hi all,

@gizmo1 I am just replying to the last thread comment. 3 weeks have passed since I've inoculated these plates and this is how they look at the moment. I am a bit worried about the possibility of them being contaminated but what really bothers me is how slow the mycelium grows. What do you guys think?

All of those plates look heavily contaminated. You should have taken transfers to new plates soon after the spores germinated.

I did the mistake to over-sprinkle many drops and with pressure into the plate, from the syringe. So basically there were droplets everywhere after I wrapped the plates.. Also the room's walls are green-turquoise and I believe they reflect a bit on the plates. Here's a photo in natural light. What do you think?

Hard to say what is what, but I wouldn't be confident transferring anything from any of those plates. I suggest you start a few new ones.

So I inoculated 2 grain jars from 2 of these dishes just to make sure and it turns out they are good.

I'd put a couple drops of green food coloring in your agar before you PC the next batch.  It helps me immensely in identifying what's growing on the plate.

That jar looks suspect to me.
