Inoculating time for millet?
    #26459250 - 01/29/20 09:49 PM (4 years, 18 days ago)

I was thinking about going with rye, but then I came across a bag of millet today, so I decided to go with that. I wasn't sure how much dry grains expand to, so I measured out a quart and half I believe hoping to make at least 4 jars to start, but I only got 3 out of it. Three being an odd number of jars, I hope I am successful!  It's in the PC right now, and then I'm going to switch it off and let it cool. However, I won't be able to get to it tomorrow morning, I will have to inoculate tomorrow afternoon after work. Will that be a problem waiting that long being greater than 12 hours? Will that affect my inoculation or it needs to be done as soon as it is done cooling? I won't be able to do it until tomorrow afternoon anyways... Also, being that I only have 3 jars, how much spore solution should I use in each jar to achieve maximum colonization? I have 12 cc's, so if I use a total of 4 cc's in each jar, should that be good enough?

You should wait at least 24 hrs after sterilizing to inoculate, but you should take most types of jars out as soon as pressure drops. Example:

I use polyfil lids, and if I leave them in the pc too long after sterilizing they get a ton of condensation on the polyfil and allow Contams to pass thru.

No matter what type of filter-lid you use, you should take it out as soon as the pressure reaches zero and the pressure indicator drops, but you shouldn’t inoculate for at least 16 hrs.

Also just prep an entire case of jars (12) at once and use 8 pounds of Miller for the whole case. Should put your jars at a little of 2/3 full each.

I would use 1/2 CC per jar if that. Less is more, because your inoculating with MS you want the least amount of genetic competition in each jar. There should be millions of spores in your syringe so the smallest drop can sometimes be the best amount to inoculate with.

They’re still in my PC from overnight. So you’re saying I should of taken the jars out and taken the foil off them?

We’re the polyfil kids? If so, yeah. Don’t leave grain jars in the PC overnight, the steam will condensate in the filter and let contaminants grow on it. Take them out as soon as the pressure drops fully.

Do I take foil off too at the same time while it's still piping hot? And where do you leave your jars to cool off and dry?

I usually leave them on my work space, If you have polyfill or SFD lids you are fine letting them air dry.

I usually leave my foil on but it’s probablygood idea to take them off. And on a shelf somewhere, as long as your filter material is proper you shouldn’t have a problem. I use a 1/4” hole stuffed with poly as my filter.
