Stay away from the middle of those two cultures. You definitely got some mold in there. You also definitely got some bacteria. Putting hot water on the dishes may have helped with the condensation, but it also created more favorable conditions for the bacteria to pop up.

On your last pictures, I'd transfer from about 4-5:30 on the first one, 4 o'clock on the second pic, and 1 o'clock on the last picture. Really be sure to focus on your sterile technique, cause you got alotta poopie in there. Remember, nothing that isnt fully sterilized can go over sterile media (this includes lids, because condensation can become bacteria bombs)

Last night did four 4 quart bags of sub for all the cyans. PC'd for 4 hours.

Hot damn, I have never had bags come out of the PC looking so good.
Perfect, vacuum sealed blocks of goodness.

Gotta say bags have been going better since I switched from 3 quarters
to 2 on the rocker.

60mm (2.3 inch) but that's the outside dimension. The lower part of the pitri is prob .25in smaller. Here's the link

Update- so I opened those weird looking pitris and found the agar was pretty firm. Maybe the myc was having a hard time getting around. This is what it looked like.



Tonight I took a bunch of projects and picked pretty much one pitri per project. Lots of pans, wild coast, British Virgin islands, and Gator clones. Plus I streaked some ksss thanks to moose Mama. Started germ plates again of Golden Halo and Rusty White. Bout time I do some work!

Side note, Pan Jams pinning and two trays of Red Down Under pinning like crazy. Also first cube mono I've ever had work is picture perfect. Pin porn coming up.

Wow over night (unless you look super close) something jumped in the T1.
             

That's just one night's difference.
What should I do?
  • 1. Leave it
    2. Take a piece of each and see what happens?  :shrug:


Edit: Aha! Once I pulled the plates out of shoeboxes they picked up the pace, including some plates with contam already!

if you mean take a piece of each growth, no, the stuff at the top is mold

take a transfer of your myc asap before that mold takes over the whole dish

I'm having alot of trouble with my plates. The myc growth is so thin and almost invisible. It's stays real thin while it colonized.
I think it's from nutes being so high. I've changed the recipe a few times and trying to do 9g agar 7-6g brf.
Potato flakes-and honey was my old one and they grew fine on that.
I've used grain soak and would that be why? I didn't cut it with water. Just agar and soak.
Should I go back to plates T1 or 2 or just transfer out of the T4 AND UP?

Progress from plates dropped 5 days ago, will be shaking in 2-3 days


last round of agar, all getting dropped to grain to sink or swim also in a few more days

Those spots do look like bacteria to me. Thank you so much! I read your response and immediately took action. I had a few agar plates ready to transfer to.

Interesting enough, I took a look at the original 4 agar plate and noticed wondered what the dark orange/brown color was at the bottom of the agar that showed up after my first extraction:

You should try taking smaller pieces of agar, the size of a grain of rice or smaller if possible and use one piece per new plate in middle of the plate. And that transfer looks dangerously close to that mold colony, he suggested 6 o clock and it looks like you went closer to 4

...full time job, wife, 8 kids, 2 dogs, 2 cats, squirrel, hampster. OMG! Only had two myself. LOL
So I'm new here but you can see the recipe I used, which has similar ingredients, but diff proportions.
4g agar, 7g potato, 50% Oat broth, 400mL. From Sagari.
And the myc loves the green food color: WATER, PROPYLENE GLYCOL, FD&C YELLOW 5, FD&C BLUE 1, AND PROPYLPARABEN,
This product has no significant nutritional value. -McCormick
       
Also, I think I've read going past T3/4 not so great.

Going past T3/4 is fine for the most part. T8 is probably the furthest I've taken one and I still didn't notice any loss in performance.
Sometimes you can't even get the culture clean in 3 or 4 transfers.

Please correct me if I'm wrong, I think I read it somewhere, maybe it was a dream, who the fuck knows but anyway. On a standard cube you can go several transfers, but if your working with a cross, like RW that going that many you may start losing traits of one. Not the it won't fruit or be vigorous, it just may not look like what you wanted. Hopefully this makes sense, and if I'm completely wrong, please tell me and this post with be deleted. I do not wanna give out any bad info where I've learned so much.

Crackatoa would know better than I but I have read a bit about this and even had it happen to an extent when I was starting out.
I had some CRS that after a few generations started making fruits with purple spores instead of the rusty spores they are known for. I have also seen discussion in the forums about Rusty White's doing the same and losing traits. It can also happen with albinos, or really any trait but more noticeable with visible traits. For the most part, I believe it's just suggested not to propagate or spread those around within the community. It's a normal process of genetic replication that this can happen, but if a variety is known for a certain trait such as a color, then you would only want to propagate the ones that show those traits unless you were trying to change that variant.

The more generations you have of these traits the more stable they become as the recessive traits become less and less likely to pair up. But I believe it is possible at any time even with stable strains four traits to be lost during reproduction in the same way that traits can be gained.
